为了鉴定和分析大别山牛背最长肌脂肪酸差异调控中的可变剪接(AS)事件,试验采用高通量转录组测序对高脂肪酸(HF)和低脂肪酸(LF)背最长肌样本进行 AS 事件分析,并筛选差异剪接基因(DSGs),对 DSGs 进行 GO 功能注释和 KEGG 信号通路富集分析,筛选参与关键 KEGG 信号通路的 DSGs 进行蛋白质-蛋白质相互作用(PPI)网络分析.结果表明:共鉴定出 26 499 个 AS 事件,其中 77.43%为外显子跳跃类型.共筛选出 682 个差异 AS 事件,涉及546 个差异剪接基因(DSGs).DSGs 主要参与代谢过程、肌肉发育及脂肪酸代谢,并富集在 AMPK、PPAR 等关键信号通路中.关键基因 UBC、SMAD2、RHOA、PI4K2B 和 TRAF6 在脂肪酸代谢调控中具有重要作用.说明差异 AS 可能通过多基因的相互作用参与调控大别山牛背肌脂肪酸的沉积.

In order to identify and analyze the alternative splicing(AS)events involved in the differential regulation of fatty acids in the longest dorsal muscle of Dabieshan cattle,high-throughput transcriptome sequencing was used to analyze AS events in longissimus dorsi samples with high fatty acid(HF)and low fatty acid(LF),and differentially spliced genes(DSGs)were screened.GO functional annotation and KEGG signaling pathway enrichment analysis were performed on DSGs,and DSGs involved in key KEGG signaling pathways were screened for protein-protein interaction(PPI)network analysis.The results showed that a total of 26,499 AS events were identified,of which 77.43%were exon skipping events.A total of 682 differential AS events were screened,involving 546 differential splicing genes(DSGs).The DSGs mainly participated in metabolic processes,muscle development,and fatty acid metabolism,and were enriched in key signaling pathways such as AMPK and PPAR.The key genes UBC,SMAD2,RHOA,PI4K2B,and TRAF6 played important roles in the regulation of fatty acid metabolism.The results indicated that differential alternative splicing might be involved in regulating the deposition of fatty acids in the longest dorsal muscle of Dabieshan cattle through the interaction of multiple genes.
