Comparative study on the effects of intranasal and ocular administration of Lactobacillus fermentum 283 extracellular vesicles and oral live bacteria on the intestinal microbiota of chickens

为了解滴鼻点眼发酵乳杆菌 283 细胞外囊泡(Evs283)与口服发酵乳杆菌 283(Bac283)发酵液对 SPF 鸡肠道菌群的影响,试验将 18 只 21 日龄 SPF 鸡随机分为 Evs283 组、Bac283 组和空白对照组,每组 6 只,Evs283 组每天滴鼻点眼 Evs283(200 μL/只),Bac283 组每天口服 Bac283 发酵液(1×108 cfu/mL,1 mL/只),空白对照组每天口服 PBS(1 mL/只),连续 3 d,1 周后对肠道菌群进行16S rDNA 测序分析,并进行 Alpha 多样性、Beta 多样性、菌群结构与差异性比较及标志菌种线性判别分析效应量(linear discriminant analysis effect size,LEfSe)分析.结 果 表 明:Evs283 组Simpson 指数和 Pielou_e 指数显著高于 Bac283 组和空白对照组(P<0.05);3 组在群落结构上呈现明显的分离趋势;Evs283 组瘤胃菌科(Ruminococcaceae)、理研菌科(Rikenellaceae)、芽殖菌属_A(Gemmiger_A)、瘤胃菌属_B(Ruminococcus_B)和真杆菌属_R(Eubacterium_R)等的相对丰度显著高于 Bac283 组和空白对照组(P<0.05);Bac283组的乳杆菌科(Lactobacillaceae)、经黏液真杆菌属_A(Blautia_A)、乳杆菌属(Lactobacillus)和粪肠球菌属(Enterenecus)等的相对丰度显著高于 Evs283 组和空白对照组(P<0.05).Evs283 组的标志菌种为毛螺菌目(Lachnospirale)、地中海杆菌属_A(Mediterraneibacter_A)和芽殖菌属_A,Bac283 组标志菌种为刺味杆菌科(Acutalibacteraceae)、粪单胞菌属(Faecimonas)和真杆菌目(Eubacteriales)等,空白对照组标志菌种为颤螺旋菌目(Oscillospirales)、颤螺菌科(Oscillospiraceae)、瘤胃菌科(Ruminococcaceae)、粪杆菌属(Faecousia)和丁酸梭菌属_A(Butyricicoccu_A),3 个组的标志菌种均不相同.说明 Evs283 在调节鸡肠道菌群结构方面较来源菌株 Bac283 具有更大潜力,尤其在增加关键益生菌种丰度上表现更为显著.

In order to understand the effects of oral administration of Lactobacillus fermentum 283(Bac283)and intranasal and ocular administration of its extracellular vesicles(Evs283)on the gut microbiota of SPF chickens,the experiment randomly divided 18 SPF chickens,21 days old,into three groups(n=6 per group):Evs283 group,Bac283 group,and the blank control group.Over three consecutive days,the groups were treated with intranasal and ocular administration of 200 μL Evs283 per chicken,oral administration of 1 mL Bac283 live bacteria(1×108 cfu/mL)per chicken,and oral administration of 1 mL PBS per chicken,respectively.After one week,16S rDNA sequencing analysis of the gut microbiota was performed,including Alpha diversity and Beta diversity,microbial composition comparisons,and linear discriminant analysis effect size(LEfSe)analysis of biomarker bacteria.The results showed that the Simpson and Pielou_e indices in Evs283 group were significantly higher than those in the Bac283 group and the blank control group(P<0.05).Three groups showed a distinct separation trend in community structure.The relative abundances of Ruminococcaceae,Rikenellaceae,Gemmiger_A,Ruminococcus_B,and Eubacterium_R in the Evs283 group were significantly higher than those in the Bac283 group and the blank control group(P<0.05).Meanwhile,the relative abundances of Lactobacillaceae,Blautia_A,Lactobacillus,and Enterococcus in the Bac283 group were significantly higher than those in the Evs283 group and the blank control group(P<0.05).Lachnospirales,Mediterraneibacter_A,and Gemmiger_A as the signature taxa for the Evs283 group,while Acutalibacteraceae,Faecimonas,and Eubacteriales were the discriminative taxon for the Bac283 group.The control group was characterized by Oscillospirales,Oscillospiraceae,Ruminococcaceae,Faecousia,and Butyricicoccus_A,demonstrating distinct microbial signatures among the three groups.These results indicate that probiotic-derived Evs283 have greater potential than Bac283 in modulating the intestinal microbiota of chickens,particularly in enhancing the abundance of key probiotic species.
