为了评估东北花猪(吉花系)的遗传特性并为其科学保种与选育提供依据,试验以 83 头东北花猪(吉花系)为研究对象,采用 PCR-限制性片段长度多态性分析(RFLP)方法对肉质及体躯发育相关基因(RYR1、MC4R、VRTN)单核苷酸多态性(SNP)位点进行多态性分析,结合生物信息学方法预测其野生型与突变型的 mRNA 二级结构和蛋白质二级结构,通过 SPSS 25.0 软件分析基因型、等位基因频率及群体 Hardy-Weinberg 平衡状态.结果表明:RYR1 基因等位基因 n 频率为 0.222,仅检测到 NN(0.554)和 Nn(0.446)基因型,突变导致 mRNA 二级结构热力学稳定性下降,蛋白 α-螺旋占比从 22.73%降至 13.64%;MC4R 基因 GA 基因型频率最高(0.554),等位基因 A、G 频率分别为 0.554和0.446,突变对 mRNA 二级结构稳定性影响轻微,蛋白质二级结构核心元件占比基本稳定;VRTN 基因等位基因 A、B频率分别为0.422 和0.578,AA、AB、BB 基因型频率分别为0.145,0.554,0.301,该位点符合 Hardy-Weinberg 平衡(P>0.05),突变型因插入 291 bp 片段导致 mRNA 二级结构稳定性显著提高,蛋白 α-螺旋占比从 47.06%升至 79.49%,其余位点均偏离 Hardy-Weinberg 平衡(P<0.05).说明东北花猪(吉花系)群体遗传选育过程需结合 MC4R、RYR1、VRTN位点的分子机制进行针对性优化.

In order to assess the genetic characteristics of Northeast Spotted pigs(Jihua breed)and provide a basis for scientific conservation and breeding,83 Northeast Spotted pigs(Jihua breed)were studied as research subjects.PCR-analysis of restriction fragment length polymorphism(RFLP)was used to analyze the polymorphisms of single nucleotide polymorphism(SNP)loci in the RYR1,MC4R and VRTN genes related to meat quality and body development.Bioinformatics methods were applied to predict the mRNA secondary structure and protein secondary structure of wild-type and mutant types.SPSS software was used to analyze genotype and allele frequencies,as well as the Hardy-Weinberg equilibrium of the population.The results showed that the frequency of allele n in the RYR1 gene was 0.222,with only NN(0.554)and Nn(0.446)genotypes detected.The mutation reduced the thermodynamic stability of the mRNA secondary structure,and the proportion of α-helices in the protein decreased from 22.73%to 13.64%.In the MC4R gene,the GA genotype had the highest frequency(0.554),with allele frequencies of 0.554 for A and 0.446 for G.The mutation had a minor effect on mRNA secondary structure stability,and the proportion of core elements in the protein secondary structure remained largely unchanged.For the VRTN gene,the frequencies of alleles A and B were 0.422 and 0.578,respectively,with genotype frequencies of 0.145 for AA,0.554 for AB,and 0.301 for BB.This locus was in Hardy-Weinberg equilibrium(P>0.05).The mutation,which involved a 291 bp insertion,significantly increased the stability of the mRNA secondary structure and raised the proportion of α-helices in the protein from 47.06%to 79.49%.All other loci deviated from Hardy-Weinberg equilibrium(P<0.05).It was suggested that the genetic breeding process of Northeast spotted pigs(Jihua breed)population should be combined with the molecular mechanism of MC4R,RYR1 and VRTN loci for targeted optimization.
