不同鳞伞菌株的分子鉴定与栽培特性研究OA

为开发鳞伞野生资源,明确菌株特性,为今后鳞伞品种选育与人工规模化生产提供理论依据.对采集自黑龙江省内不同地区的10株鳞伞类似菌株开展菌丝生长特性、菌丝显微结构、孢子形态特征、rDNA ITS序列分析鉴定以及栽培性状研究.结果表明:供试菌株菌落均呈圆形、具同心轮纹,菌丝浓密且边缘整齐,均具典型锁状联合,可分为粗壮型与纤细型,菌落颜色与生长速率差异显著;结合ITS序列比对与系统发育分析,6 株(HS-1、HS-2、HS-3、HS-8、HS-9、HS-10)鉴定为多脂鳞伞(Pholiota adiposa),4 株(HS-4、HS-5、HS-6、HS-7)鉴定为柠檬鳞伞(Pholiota limonella).20 ℃、90%湿度条件下,4 株菌株(HS-1、HS-3、HS-7、HS-9)成功出菇,子实体形态差异显著,且出菇菌株孢子成熟度高、活性良好,其中HS-1、HS-9综合农艺性状较优.

In order to develop wild Pholiota species resources and clarify strain characteristics,providing theoretical basis for future variety breeding and artificial large-scale production,research was conducted on 10 similar Pholiota strains collected from different regions of Heilongjiang Province.The study investigated mycelial growth characteristics,mycelial microstructure,spore morphological features,rDNA ITS sequence analysis and identification,as well as cultivation traits.Results demonstrated that all tested strains exhibited circular colonies with concentric wheel patterns,dense mycelium with uniform margins,and typical lock-like junctions.The strains could be classified into robust and slender types,showing significant differences in colony color and growth rate.Through ITS sequence alignment and phylogenetic analysis,six strains(HS-1,HS-2,HS-3,HS-8,HS-9,HS-10)were identified as Pholiota adiposa,while four strains(HS-4,HS-5,HS-6,HS-7)were identified as Pholiota limonella.Under conditions of 20℃ and 90%humidity,four strains(HS-1,HS-3,HS-7,HS-9)successfully produced fruiting bodies with significant morphological variations.Among them,HS-1 and HS-9 exhibited superior comprehensive agronomic traits,and all four fruiting strains demonstrated high spore maturity and good viability.
