目的:探讨六位地黄丸促进多发性硬化髓鞘再生的作用和机制.方法:60只C57BL/6J小鼠,随机分为正常组、模型组、芬戈莫德组和六味地黄丸低、中、高剂量组,每组10只.模型组和给药组小鼠通过喂养含0.2%双环己酮草酰二腙的饲料5周来诱导脱髓鞘,从第6周开始恢复正常饮食,同时给药组小鼠进行灌胃给药,直至第 8周末取材.经快蓝染色观察有髓面积变化;免疫组化测定髓鞘碱性蛋白(myelin basic protein,MBP)的表达,观察髓鞘再生情况;免疫荧光法测定星形胶质细胞A1/A2极化标志物,观察该方对A1/A2极化的调节作用;ELISA 法检测细胞因子 IL-1α、TNF-α的表达;Western blot检测小鼠脑组织中MBP、C3、S100A10、p65和p-p65蛋白的表达.结果:六味地黄丸灌胃后,小鼠脑组织的髓鞘LFB染色增加,MBP表达增多(P<0.01),NF-κB信号通路关键蛋白p65、p-p65及相关细胞因子TNF-α和IL-1α的表达减少(P<0.05),AST向A1方向极化减少,C3表达减少;AST向A2方向极化增多,S100A10表达增多.结论:六味地黄丸可能通过调控NF-κB信号通路及相关细胞因子,明显减轻神经炎症,介导A1/A2极化,促进多发性硬化髓鞘再生.

Objective:To explore the efficacy and mechanism of Liuwei Dihuang Pills in promoting remyelination in multiple sclerosis.Methods:A total of sixty C57BL/6J mice were randomly divided into the normal control group,the model group,the Fingolimod group and the Liuwei Dihuang Pill low-dose,medium-dose and high-dose groups,with 10 mice in each group.The mice in the model group and the drug group were fed with a diet containing 0.2%dicyclohexanone oxalyl dihydrazone for 5 weeks to induce demyelination.From the 6th week onwards,the normal diet was resumed.Meanwhile,the mice in the treatment group were given intragastric administration of the drug until the end of the 8th week when samples were collected.The luxol fast blue staining observation myelinated area of change;Immunohistochemical determination of the expression of myelin basic protein(MBP),observe the myelin regeneration;Immunofluorescence determination of astrocyte A1/A2 polarization markers,observe the party works for the regulation of A1/A2 polarization;ELISA method to detect the corresponding cytokine IL-1α and TNF-α.Western blot was used to detect the expression of MBP,C3,S100A10,p65 and p-p65 proteins in mice brain tissues.Results:Af-ter gavage administration of Liuwei Dihuang Pill,the myelin sheath LFB staining and MBP expression of mouse brain tissue in-creased(P<0.01).The expressions of key proteins p65 and p-p65 and related cytokines TNF-α and IL-1α in the NF-κB signaling pathway were decreased(P<0.05).AST polarization towards A1 is reduced,and C3 expression is reduced.The polarization of AST in the direction of A2 increased,and the expression of S100A10 increased.Conclusion:Liuwei Dihuang Pill may significant-ly alleviate neuroinflammation,mediate A1/A2 polarization,and promote remyelination in multiple sclerosis by regulating the NF-κB signaling pathway and related cytokines.
