秋水仙素对天贵卷瓣兰多倍体的诱导效应OA

Induction effect of colchicine on polyploids of Bulbophyllum tianguii

[目的]利用秋水仙素诱导获得天贵卷瓣兰多倍体,为其种质创新与育种提供技术与方法.[方法]采用不同质量浓度秋水仙素作为诱变剂,通过浸泡法与混培法处理天贵卷瓣兰组培苗丛生芽,并采取流式细胞术、形态学观察及根尖染色体计数等方法鉴定诱变效果.[结果]经浸泡法和混培法诱导获得大量嵌合体,浸泡法获得的四倍体更多,其中 150 mg·L-1秋水仙素浸泡 12 h 处理诱导率最高(6.67%),但嵌合率 38.33%、死亡率 36.67%.多倍体植株与二倍体相比,其DNA 荧光强度吸收峰值存在明显差别.在叶片形态和气孔形态对比中,四倍体植株叶片颜色深绿,叶长、叶宽、保卫细胞长度及宽度等均大于二倍体植株;但气孔密度(每视野 14 个)显著小于二倍体植株(每视野 24 个).根尖细胞染色体计数显示,二倍体植株的染色体数目为:2n=2x≈44 条,四倍体植株染色体数目为:2n=4x≈88 条.[结论]秋水仙素可成功诱导天贵卷瓣兰产生多倍体,诱导效果受秋水仙素质量浓度和处理时间的影响,多倍体植株表型特征优于二倍体.

[Purpose]Colchicine was used to induce polyploidy in Bulbophyllum tianguii,providing reliable techniques and meth-ods for germplasm innovation and genetic breeding.[Method]Different concentrations of colchicine were used as mutagens.Clus-tered buds of in vitro B.tianguii plantlets were treated via the soaking and mixed-culture methods.The mutagenic effects were sys-tematically evaluated through flow cytometry,morphological observation,and root-tip cell chromosome counting.[Result]Numer-ous chimeras were induced through both the soaking and mixed-culture methods.The soaking method yielded more tetraploids.The treatment with the highest induction rate(6.67%)was soaking in 150 mg·L-1 colchicine for 12 h,while it showed a chimera rate of 38.33%and a mortality rate of 36.67%.Successfully induced polyploid plants showed significant differences in DNA fluorescence intensity absorption peaks compared with diploids.In terms of leaf and stomatal morphology,tetraploid plants had darker green leav-es,longer and wider leaves,and larger gurd cells(both in length and width)compared with diploid plants.However,their stomatal density(14 per field of view)was significantly lower than that of diploid plants(24 per field of view).Root-tip cell chromosome counting showed that the chromosome number was 2n=2x≈44 in diploid plants and 2n=4x≈88 in tetraploid plants.[Conclusion]Colchicine can successfully induce polyploidy in B.tianguii.The induction effects are influenced by colchicine concentration and treatment duration.The polyploid plants exhibit superior phenotypic traits compared with diploids.
