甘蔗耐寒相关基因ScERF118的克隆与表达分析OA
Cloning and expression analysis of the cold tolerance-associated gene ScERF118 in sugarcane
[目的]克隆与鉴定甘蔗耐寒相关 ERF 转录因子基因,为甘蔗抗寒遗传改良提供候选靶点.[方法]基于本课题组前期通过群体连锁定位获得的甘蔗耐寒相关 ERF 转录本,以粤糖 93-159 为材料,采用同源克隆方法获得 ScERF118 基因cDNA序列;利用生物信息学方法分析其编码蛋白的理化性质、系统进化、保守结构域、启动子区顺式作用元件;结合转录组数据与 RT-qPCR 技术,分析该基因在不同胁迫条件下的表达特征;通过原生质体瞬时转化系统观察 ScERF118 蛋白的亚细胞定位.[结果]ScERF118 基因 cDNA 序列长度为 1 529 bp,含有 1 个 1 053 bp 的开放阅读框,编码 350 个氨基酸.该编码蛋白被预测为不稳定的酸性亲水性蛋白,不含信号肽和跨膜结构域.亚细胞定位结果显示,ScERF118 定位于细胞核.系统进化分析表明,ScERF118 属于 ERF 亚家族 B-1 分支,其 AP2 结构域第 14 位丙氨酸和第 19 位天冬酰胺在多个物种中较为保守.ScERF118 同源基因启动子区富含脱落酸、乙烯、茉莉酸甲酯响应元件及 DRE、LTR 等低温和干旱胁迫响应元件.转录组分析表明,该基因在甘蔗各组织中表达量极低,但可被低温、干旱及黑穗病菌胁迫诱导表达.RT-qPCR 验证结果表明,ScERF118 基因在低温胁迫下普遍被诱导上调表达,且在抗寒品种中的响应强度高于敏感品种.[结论]ScERF118 是一个定位于细胞核、响应低温等多种逆境信号的非组成型表达转录因子基因,可能在低温胁迫应答中发挥调控功能.本研究为该基因的功能解析奠定了基础,并为甘蔗抗寒分子育种提供了候选基因资源.
[Purpose]This study cloned and characterized an ERF transcription factor gene associated with cold tolerance in sugar-cane,aiming to provide a candidate target for genetic improvement of sugarcane for cold tolerance.[Method]On the basis of a sug-arcane cold tolerance-associated ERF transcript previously identified by our population-based linkage mapping,the cDNA sequence of ScERF118 was obtained from the sugarcane cultivar YT93-159 via homology-based cloning.The physicochemical properties,phy-logenetic relationship,conserved domains,and promoter cis-acting elements(CREs)of the protein encoded by ScERF118 were ana-lyzed by bioinformatics tools.Expression profiles of the gene under various stress conditions were examined by transcriptomic data and RT-qPCR.Subcellular localization of ScERF118 was determined via a protoplast transient expression system.[Result]The cD-NA sequence of ScERF118 was 1 529 bp in length and contained an open reading frame of 1 053 bp,encoding 350 amino acid resi-dues.The predicted protein was an unstable,acidic,and hydrophilic protein without a signal peptide or transmembrane domain.Subcellular localization results showed that ScERF118 was localized to the nucleus.Phylogenetic analysis revealed that ScERF118 belonged to the B-1 subgroup of the ERF subfamily.The 14th alanine(A)and 19th asparagine(N)residues within the AP2 do-main were conserved across multiple species.The promoter region of the ScERF118 homologous gene contained abundant CREs re-sponsive toabscisic acid,ethylene,and methyl jasmonate,as well as low-temperature and drought stress-responsive elements such as DRE and LTR.Transcriptomic data showed that ScERF118 expression was very low across various sugarcane tissue samples but in-duced by low temperature,drought,and smut pathogen stresses.RT-qPCR validation demonstrated that the expression of ScERF118 was generally upregulated under low temperature stress,with a stronger response observed in cold-tolerant varieties than in cold-sen-sitive ones.[Conclusion]ScERF118 is a nuclear-localized,non-constitutively expressed transcription factor responsive to low tem-perature and other stress signals,potentially playing a regulatory role in the response to low temperature stress.This study lays a foundation for functional characterization of ScERF118 and provides a candidate target for cold tolerance breeding in sugarcane.
