Advances in Nanoneedle-based Live-Cell Biomarker Sensing and Analysis
传统的活细胞分子检测技术存在分子扰动和探针毒性等问题.纳米材料凭借其良好的生物相容性,为活细胞检测提供了新途径.本文聚焦于纳米针阵列的特殊结构,系统梳理了其在活细胞标志物分析中的原理、应用与前景;详细总结了纳米针阵列的材料构成、形貌特征、表面修饰策略以及分子捕获与信号转导的工作机制;深入分析了其对核糖核酸(Ribonucleic acid,RNA)、蛋白质及代谢物的原位检测能力.此外,本文还探讨了该技术在时空维度整合分析方面的优势,并剖析了临床转化面临的挑战,为单细胞异质性解析、疾病机制探索及药物响应规律研究提供了方法学基础.
Traditional live-cell molecular detection techniques are often impeded by the challenges such as molecular perturbation and probe-induced toxicity.Leveraging their superior biocompatibility,nanomaterials offer a novel paradigm for live-cell interrogation.This review focused on nanoneedle arrays,systematically outlining their principles,applications,and prospects for biomarker analysis.Material compositions,morphological features,and surface functionalization strategies of these arrays were elucidated in detail,alongside the mechanisms governing molecular capture and signal transduction.Specifically,the capability of nanoneedle arrays for in situ detection of RNA,proteins,and metabolites were critically evaluated.Furthermore,the technology's advantages in spatiotemporal analysis were highlighted and the hurdles facing clinical translation were dissected.Ultimately,this paper aimed to establish a robust methodological foundation for deciphering single-cell heterogeneity,exploring disease mechanisms,and elucidating drug-response profiles.
