为探究长期施肥对黑土氨氧化微生物丰度和硝化作用的影响,依托东北黑土长期定位施肥试验平台(40 a),以不施氮肥、单施氮肥、单施磷肥和氮磷肥配施 4 种处理的黑土为研究对象,采用定量 PCR技术连续 3 年(2021-2023 年)分析氨氧化微生物丰度的年际变化,并通过氯酸盐抑制法测定土壤硝化潜势.结果表明,氮肥或氮磷配施均显著降低黑土氨氧化古菌、完全氨氧化菌分支 B 的丰度及土壤硝化潜势,显著提高氨氧化细菌、完全氨氧化菌分支 A 丰度及 NO3--N 含量.单施磷肥对氨氧化微生物丰度无显著影响,但显著提高土壤硝化潜势.相关分析显示,氨氧化古菌丰度为影响土壤硝化潜势的主要生物因子,而土壤 pH为主要环境因子.综上,长期施肥通过改变氨氧化微生物丰度与土壤 pH,进而影响黑土硝化作用.

This study investigates the effects of long-term fertilization on the abundance of am-monia-oxidizing microorganisms and potential nitrification rate in black soil,based on a long-term(40 years)fertilization experiment in Northeast China.Soil samples were collected from four treat-ments,including no nitrogen fertilizer(CK),nitrogen fertilizer only(N),phosphorus fertilizer only(P),and combined nitrogen-phosphorus fertilizers(NP),over three consecutive years(2021-2023).The abundance of ammonia-oxidizing microorganisms was quantified via real-time quantitative PCR,and the soil potential nitrification rate was measured using the chlorate inhibition method.The results showed that both N and NP treatments significantly decreased the abundance of ammonia-oxi-dizing archaea,comammox Nitrospira Clade B,and the soil potential nitrification rate,while signifi-cantly increasing the abundance of ammonia-oxidizing bacteria,comammox Nitrospira Clade A,and soil NO3--N content.The P treatment had no significant effect on the abundance of ammonia-oxidizing microorganisms,but significantly increased the soil potential nitrification rate.Correlation analysis in-dicated that the abundance of ammonia-oxidizing archaea was the dominant biotic factor influencing the soil potential nitrification rate,while soil pH was the main environmental factor.In conclusion,long-term fertilization regulates soil nitrification in black soil by altering the abundance of ammonia-oxidizing microorganisms and soil pH.
