嗜水气单胞菌分离株的鉴定及MLST分型检测OA
Identification and MLST Typing Detection of Aeromonas hydrophila Isolates
为调查镇江市淡水鱼源嗜水气单胞菌的流行和分类情况,在分离自该市若干养殖渔场的74 株病原菌中纯化得到嗜水气单胞菌,并进行形态学观察、gyrB 基因序列分析以及多位点序列分型(Multilocus sequence typing,MLST)检测.结果显示:嗜水气单胞菌单菌落在 RS 平板上呈淡黄色,圆形、光滑、边缘整齐,革兰氏染色后镜检可见两端钝圆的红色杆菌;经分离株gyrB 基因序列比对分析后鉴定出气单胞菌 40 株,其中 12 株初步确定为嗜水气单胞菌;进一步构建了基于 gyrB 基因的气单胞菌分离株系统发育树,确定其中 8 株分离株为嗜水气单胞菌.对嗜水气单胞菌管家基因 gyrB、groL、gltA、metG、ppsA、recA 进行 PCR 扩增和序列分析,鉴定出ST251 型嗜水气单胞菌 2 株,ST378 型 3 株,ST469、ST361、ST1075 型各 1 株.研究结果表明该地区存在 ST251 型强致病性嗜水气单胞菌流行的潜在风险,危害当地水产养殖业的绿色高质量发展.
In order to investigate the prevalence and classification of Aeromonas hydrophila from the freshwater fish in Zhenjiang City,A.hydrophila was purified from 74 strains of pathogenic bacteria isolated from several aquaculture farms in the city,and then the morphological observation,gyrB gene sequence analysis and multilocus sequence typing(MLST)were conducted.The results showed that A.hydrophila appeared as light yellow,round,smooth,and neatly edged colonies on RS agar plates and red,rod-shaped bacteria with blunt ends by microscopic examination after Gram staining.After conducting the sequence alignment analysis on the gyrB gene of isolated strains,40 strains of Aeromonas were identified,of which 12 were preliminarily determined to be A.hydrophila.The phylogenetic tree of Aeromonas isolates based on the gyrB gene was further constructed,finally identifying eight isolates as A.hydrophila.The PCR amplification and sequence analysis were performed on the housekeeping genes gyrB,groL,gltA,metG,ppsA,and recA of A.hydrophila.Two strains of A.hydrophila type ST251,three strains of type ST378,and one strain each of types ST469,ST361,and ST1075 were identified.The results showed that there was a potential risk of the prevalence of ST251 type of highly pathogenic A.hydrophila in the region,which endangered the green and high-quality development of the local aquaculture industry.
