中国南瓜生长调节因子的基因鉴定及表达分析OA
Gene Identification and Expression Analysis of Growth-regulating Factors in Cucurbita moschata
生长调节因子(Growth-regulating Factors,GRFs)是一种植物特异性的转录因子,已被证实参与叶片生长、花器官发育及植物激素信号传导等多个生物学过程.利用生物信息学方法对南瓜 GRF 基因家族成员进行鉴定,分析其编码蛋白的理化性质、染色体定位、基因结构、进化关系、共线性及启动子内的顺式元件,并结合转录组分析南瓜 GRF 基因家族在不同组织中的表达模式.结果表明:在南瓜基因组中鉴定出 14 个 GRF 基因家族成员,其分布在 11 条不同的染色体上.通过系统发育分析,可以将 CmoGRF 基因家族成员归属到 4 个亚族.在 CmoGRF 基因家族中鉴定出 11 对共线性基因,但未发现串联重复基因;根据种间共线性分析,9 个CmoGRF 基因在拟南芥中具有共线基因对,5 个 CmoGRF 基因在水稻中具有相应的同源基因.在 CmoGRFs 启动子中,筛选出 5 类与植物激素反应相关的元件、4 类代谢分化调控元件、4 类蛋白结合位点和 3 类参与防御和应激反应的顺式作用元件.14 个 CmoGRFs 基因在不同组织中的相对表达差异显著.CmoGRF1、CmoGRF12 和 CmoGRF13 基因的表达模式分析发现,CmoGRF1 基因在 6-苄氨基嘌呤(6-BA)、茉莉酸甲酯和玉米素诱导下高表达;CmoGRF12 在茉莉酸甲酯诱导下高表达;CmoGRF13 在脱落酸和茉莉酸甲酯处理下表达量显著提高.综上所述,研究结果为深入解析南瓜 GRF 基因家族的功能特性和分子机制奠定了基础,并为南瓜的育种提供了理论基础和新的基因资源.
Growth-regulating factors(GRFs),a type of plant-specific transcription factors,have been demonstrated to be involved in various biological processes,including leaf growth,floral organ development,and phytohormone signal transduction.In this study,GRF gene family members in Cucurbita moschata were identified by using the bioinformatic methods.The physicochemical properties,chromosomal localization,gene structure,evolutionary relationship,collinearity and cis-acting elements within the promoter regions were analyzed.The expression patterns of CmoGRF genes across different tissues were further analyzed in combination with transcriptome analysis.The results showed that a total of 14 CmoGRF genes family members were identified in the pumpkin genome,which were distributed across 11 different chromosomes.Through the phylogenetic analysis,the members of the CmoGRF gene family could be classified into four subgroups.Eleven pairs of syntenic genes were detected within the CmoGRF family,although no tandem duplicated genes were observed.The interspecies collinearity analysis revealed that nine CmoGRF genes had syntenic counterparts in Arabidopsis thaliana,while five CmoGRF genes were homologous to the genes in Oryza sativa.Within the promoters of CmoGRFs,five types of elements related to plant hormone response,four types of metabolic differentiation regulatory elements,four types of protein-binding sites,and three types of cis-acting elements associated with the defense and stress responses were screened.The relative expression levels of the 14 CmoGRFs genes varied significantly across different tissues.The expression pattern analysis of CmoGRF1,CmoGRF12,and CmoGRF13 revealed that,the CmoGRF1 gene was highly expressed under the induction of 6-benzylaminopurine(6BA),methyl jasmonate and zeatin.The CmoGRF12 gene was highly expressed under the induction of methyl jasmonate.The expression of the CmoGRF13 gene was significantly increased under the treatments with abscisic acid and methyl jasmonate.In summary,the results laid a foundation for further analysis on the functional characteristics and molecular mechanisms of the GRF gene family in Cucurbita moschaa,providing a theoretical foundation and new genetic resources for the breeding of pumpkin.
