冬眠期和非冬眠期雄性达乌尔黄鼠食欲相关基因表达及激素水平的变化OA
The variation of appetite genes expression and hormones levels in hibernating and non-hibernating male daurian ground squirrels
[目的]达乌尔黄鼠(Spermophilus dauricus)是栖息于干草原和荒漠草原的一种冬眠动物,其摄食行为具有独特的季节变化,研究旨在探究冬眠期与非冬眠期达乌尔黄鼠食欲调节的可能机制.[方法]通过RT-qPCR法检测夏季(活跃期,7月)、冬季(冬眠期,12月)和春季(出眠期,3月底)雄性达乌尔黄鼠胃中神经元型一氧化氮合酶(nNOS)、神经肽Y(NPY)、血管活性肠肽(VIP)、毒蕈碱型胆碱受体M2(CHRM2)和胃饥饿素(GHRL)基因的表达以及海马nNOS和瘦素受体(LepR)的相对基因表达.通过酶联免疫方法检测血清八肽胆囊收缩素(CCK-8)的浓度.[结果]达乌尔黄鼠冬季胃中nNOS和VIP基因的表达显著低于春季或夏季,CHRM2的表达显著高于春季和夏季;冬季和春季的GHRL表达显著低于夏季;冬季NPY表达较春季和夏季有降低趋势,但差异不显著.冬季和春季海马的nNOS和LepR基因的表达显著低于夏季.冬季血清CCK-8浓度低于春季和夏季.[结论]nNOS,CHRM2,VIP和GHRL基因可能通过协同调节胃运动参与调节达乌尔黄鼠不同季节的摄食变化,海马nNOS和LepR基因及CCK也参与了摄食行为的季节节律.
[Objective]The daurian ground squirrel(Spermophilus dauricus),a hibernating rodent inhabiting arid and semi-arid steppes,exhibits pronounced seasonal variation in feeding behavior.This study aimed to investi-gate the molecular mechanisms underlying appetite regulation during hibernation and active periods.[Method]Gene expression levels of neuronal nitric oxide synthase(nNOS),neuropeptide Y(NPY),vasoactive intestinal peptide(VIP),muscarinic acetylcholine receptor M2(CHRM2),and ghrelin(GHRL)were measured in gastric tissues us-ing RT-qPCR.Hippocampal expression of nNOS and leptin receptor(LepR)was also analyzed in male daurian ground squirrels during summer(active period,July),winter(hibernating period,December),and spring(arousal pe-riod,late March).Serum cholecystokinin-8(CCK-8)levels were assessed by ELISA.[Result]Gastric expres-sion of nNOS and VIP was significantly lower in winter than in spring or summer,whereas CHRM2 expression was significantly higher in winter.GHRL expression in winter and spring was significantly lower than in summer.NPY ex-pression showed a decreasing trend in winter,although the difference was not statistically significant.The nNOS and LepR expression in the hippocampus were significantly lower in winter and spring than in summer.Serum CCK-8 concentrations were also lower in winter than in summer and spring.[Conclusion]The genes nNOS,CHRM2,VIP,and GHRL may cooperatively regulate gastric motility,thereby modulating seasonal feeding variations in daurian ground squirrels.Additionally,hippocampal nNOS and LepR genes,along with the hormone CCK,contribute to the seasonal rhythmicity of feeding behavior.These findings provide insight into the adaptive mechanisms underlying sea-sonal appetite regulation in hibernating mammals.
