目的 基于3 种动物模型药效评价体系观察柴芩颗粒对动物急性炎症的抗炎作用及机制.方法 通过构建二甲苯致小鼠耳肿胀、冰醋酸致小鼠腹腔毛细血管通透性增加及角叉菜胶致大鼠足趾肿胀 3 种炎症模型,观察柴芩颗粒的抗炎作用;采用酶联免疫吸附法(ELISA)检测角叉菜胶致炎大鼠血清及足趾肿胀组织中 IL-6、IL-17、IL-1β、IL-18、NLRP3 炎症小体及 Caspase 1 的含量.结果 与对照组比较,柴芩颗粒中、高剂量组均可抑制二甲苯致小鼠耳肿胀(P<0.01,P<0.05);柴芩颗粒中剂量组可明显抑制醋酸致小鼠腹腔毛细血管通透性增加(P<0.05);柴芩颗粒高、中、低剂量组可不同程度降低大鼠血清中炎症因子IL-6、IL-17、IL-1β 含量(P<0.05,P<0.01,P<0.001),同时降低足趾组织中 IL-18、IL-1β、Caspase l、NLRP3 的含量(P<0.05,P<0.01,P<0.001).结论 柴芩颗粒对急性炎症有抑制作用,可通过抑制 NLRP3 炎症小体信号通路、调控炎性因子 IL-1β 和 IL-18 释放,降低血清中炎症因子 IL-6、IL-17、IL-1β 水平以缓解炎症反应.

Objective To investigate the anti-inflammatory effect and mechanisms of Chaiqin Granules on acute inflammation in animals based on a pharmacodynamic evaluation system with three animal models.Methods Three inflammatory models were established,including xylene-induced ear edema in mice,acetic acid-induced increased peritoneal capillary permeability in mice,and carrageenan-induced paw edema in rats,to observe the anti-inflam-matory effect of Chaiqin Granules.Enzyme-linked immunosorbent assay(ELISA)was employed to detect the contents of IL-6,IL-17,IL-1β,IL-18,NLRP3 inflammasome and Caspase 1 in the serum and paw edema tissues of carrageenan-induced inflammatory rats.Results Compared with control group,medium and high-dose groups of Chaiqin Granules both inhibited xylene-induced ear edema in mice(P<0.01,P<0.05).Medium-dose group of Chaiqin Granules significantly suppressed acetic acid-induced increase in peritoneal capillary permeability in mice(P<0.05).High,medium and low-dose groups of Chaiqin Granules reduced the contents of inflammatory factors IL-6,IL-17 and IL-1β in rat serum to varying degrees(P<0.05,P<0.01,P<0.001),and simultaneously decreased the contents of IL-18,IL-1β,Caspase 1 and NLRP3 in paw tissues(P<0.05,P<0.01,P<0.001).Conclusion Chaiqin Granules have an inhibitory effect on acute inflammation.They can alleviate inflammatory responses by inhibiting the NLRP3 inflammasome signaling pathway,regulating the release of inflammatory factors IL-1β and IL-18,and reducing the levels of inflammatory factors IL-6,IL-17 and IL-1β in serum.
