五味子酚对小鼠肝癌H22细胞移植瘤生长的影响OA
Effect of Schisanhenol on the Growth of Transplanted H22 Hepatoma Cells in Mice
目的 探究五味子酚(schisanhenol,SAL)对小鼠肝癌 H22 细胞移植瘤生长的影响.方法 构建小鼠肝癌H22 细胞移植瘤模型,待移植瘤形成后,将15 只 C57BL/6 小鼠分为模型组、SAL 10 mg/kg 组、SAL 20 mg/kg 组,每组5 只.采用游标卡尺检测小鼠移植瘤体积,计算抑瘤率;通过苏木精-伊红染色和免疫组化染色法,分别检测 SAL 对小鼠肝癌 H22 细胞移植瘤组织形态及移植瘤组织中 Ki-67 蛋白表达的影响;采用 Western blot 法检测小鼠移植瘤组织中 p-STAT3(Tyr705)、Bcl-2 和 Cyclin D1 蛋白的表达.结果 与模型组比较,SAL 10、20 mg/kg组均显著抑制小鼠肝癌 H22 细胞移植瘤体积(P<0.05),且呈剂量依赖性.免疫组化染色结果显示,与模型组比较,SAL 10、20 mg/kg 组均可抑制小鼠肝癌 H22 细胞移植瘤组织中 Ki-67 蛋白表达.Western blot 结果显示,与模型组比较,SAL 10、20 mg/kg 组均显著抑制 p-STAT3(Tyr705)、Bcl-2 和 Cyclin D1 蛋白表达(P<0.01),且呈剂量依赖性.结论 五味子酚抑制小鼠肝癌 H22 细胞移植瘤生长,其机制可能与抑制 p-STAT3(Tyr705)和下游凋亡基因 Bcl-2表达、促进 Cyclin D1 表达有关.
Objective To investigate the effect of schisanhenol(SAL)on the growth of transplanted H22 hepa-toma cells in mice.Methods A mouse model of transplanted H22 hepatoma cells was established.After the formation of transplanted tumors,15 C57BL/6 mice were divided into model group,SAL 10 mg/kg group,and SAL 20 mg/kg group,with 5 mice in each group.The volume of transplanted tumors in mice was measured using a vernier caliper,and the tumor inhibition rate was calculated.Hematoxylin-eosin staining and immunohistochemical staining were used to detect the effects of SAL on the histomorphology of transplanted H22 hepatoma tissues and the expression of Ki-67 protein in transplanted tumor tissues,respectively.Western blot was employed to determine the expression levels of p-STAT3(Tyr705),Bcl-2,and Cyclin D1 proteins in mice transplanted tumor tissues.Results Compared with model group,both SAL 10 and 20 mg/kg groups significantly inhibited the volume of transplanted H22 hepatoma cells in mice(P<0.05)in a dose-dependent manner.The results of immunohisto-chemical staining showed that compared with model group,both SAL 10 and 20 mg/kg groups could inhibit the expression of Ki-67 protein in the transplanted H22 hepatoma tissues of mice.Western blot results demonstrated that compared with model group,both SAL 10 and 20 mg/kg groups significantly inhibited the expression of p-STAT3(Tyr705),Bcl-2,and Cyclin D1 proteins(P<0.01)in a dose-dependent manner.Conclusion Schisan-henol inhibits the growth of transplanted H22 hepatoma cells in mice,and its mechanism may be related to the in-hibition of p-STAT3(Tyr705)and the expression of downstream apoptotic gene Bcl-2,as well as the promotion of Cyclin D1 expression.
