不同产地生地黄主要成分与抗氧化活性评价OA

分析不同产地地黄主要化学成分及体外抗氧化活性差异,为其药材质量评价及资源合理开发利用提供依据.采用HPLC-PDA法测定16批地黄中梓醇、地黄苷D、毛蕊花糖苷及松果菊苷的含量;通过DPPH和ABTS自由基清除实验评价其体外抗氧化活性,结合聚类分析、主成分分析及Pearson相关性分析探讨化学成分与抗氧化活性的关联.结果显示不同产地地黄样品化学成分差异显著.梓醇含量最高的为新疆从山西临汾引种样品(31.021 mg/g),地黄苷D含量最高的为河南焦作样品(5.633 mg/g),松果菊苷和毛蕊花糖苷含量最高的分别为新疆从山西曲沃县引种样品(0.509 mg/g)及新疆从山西保焉引种样品(1.352 mg/g).新疆从山西曲沃县引种样品抗氧化活性最强(DPPH IC50 为 8.97 mg/mL,ABTS IC50 为5.29 mg/mL).相关性分析表明,松果菊苷含量与DPPH自由基清除能力(IC50)呈显著负相关(P<0.05),提示其为关键抗氧化成分.新疆引种地黄在特定化学成分积累及综合抗氧化活性方面表现出独特优势,本研究为地黄的质量评价、产地优选及关键质量标志物的确定提供了科学依据.

To investigate the differences in main chemical components and in vitro antioxidant activity of Rehmannia glutinosa Libosch.from different geographical origins,and to provide a basis for the quality evaluation and rational utiliza-tion of its medicinal resources.The contents of catalpol,rehmannioside D,verbascoside,and echinacoside in 16 batches of Rehmannia glutinosa Libosch.were determined by HPLC-PDA.The antioxidant activities were evaluated by DPPH and ABTS free radical scavenging assays.The relationship between chemical components and antioxidant activity was ex-plored by combining cluster analysis(HCA),principal component analysis(PCA),and Pearson correlation analysis.Re-sults showed that there were significant differences in the chemical composition of samples from different origins.The highest content of catalpol(31.021 mg/g)was found in samples introduced from Linfen,Shanxi to Xinjiang.The high-est content of rehmannioside D(5.633 mg/g)was in samples from Jiaozuo,Henan.The highest contents of echinacoside(0.509 mg/g)and verbascoside(1.352 mg/g)were in samples introduced from Quwo County and Baoyan,Shanxi to Xinjiang,respectively.Samples introduced from Quwo County,Shanxi to Xinjiang exhibited the strongest antioxidant ac-tivity(DPPH IC50 8.97 mg/mL,ABTS IC50 5.29 mg/mL).Correlation analysis indicated that the content of echinaco-side was significantly negatively correlated with the DPPH IC50 value(P<0.05),suggesting it as a key antioxidant com-ponent.In summary,the introduced Rehmannia glutinosa Libosch.in Xinjiang exhibits unique advantages in terms of specific chemical component accumulation and comprehensive antioxidant activity.This study provides a scientific basis for the quality evaluation,origin selection,and identification of key quality markers of Rehmannia glutinosa Libosch.
