低温高分子糖降解菌的筛选及产酶条件研究OA
Screening of Low Temperature Polymeric Sugar Degrading Bacteria and Study on Enzyme Production Conditions
从北方农业土壤中分离筛选出一株产淀粉酶和纤维素酶的低温菌,通过16S rDNA 序列比对分析,该菌为Glutamicibacter arilaiten-sis.对该菌产淀粉酶和纤维素酶的培养条件进行初步研究,结果表明:该菌产淀粉酶和纤维素酶的最适温度均为10℃,最适pH 均为7;淀粉酶的最佳产酶时间为 48 h,此时粗酶液淀粉酶活力最高,为 12.744 U/mL;产纤维素酶的最佳培养时间为 96 h,酶活力为4.882 U/mL.该菌株在10℃低温条件下可同时产生淀粉酶和纤维素酶,在北方地区农牧业废物、废水处理中具有一定的研究价值和应用前景.
A low temperature bacterium producing amylase and cellulase was isolated and screened from agricultural soil in northern China.After 16S rDNA sequence alignment analysis,it was determined that the bacterium is Glutamicibacter arilaitensis.Preliminary study was con-ducted on the cultivation conditions for producing amylase and cellulase by this bacterium.The results showed that the optimal temperature for the production of two enzymes by this bacterium was 10℃and the optimal pH was 7.After 48 hours of cultivation,the highest amylase activi-ty was observed in the crude enzyme solution,which was 12.744 U/mL.While the optimal fermentation time for producing cellulase was 96 hours,with a highest enzyme activity of 4.882 U/mL.Due to its ability to produce both amylase and cellulase under low temperature condi-tions of 10℃,this strain has certain research value and application prospects in the treatment of agricultural and livestock waste and wastewater in northern regions.
