自选小麦新品种(系)的高分子量麦谷蛋白亚基组成分析OA
Analysis on the Composition of High-molecular-weight Gluten Subunits in the New Wheat Lines Self-selected
高分子量麦谷蛋白亚基(HMW-GS)是影响小麦品质的重要因素,对其组成进行分析与鉴定是选育优质新品种的重要内容.以自主选育的42个小麦新品种(系)为材料,在综合农艺学鉴定基础上,采用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)技术和分子标记等方法,对其HMW-GS组成和品质相关性状进行鉴定.结果表明,42份小麦新品种(系)中,HMW-GS共出现了9种亚基类型.在Glu-A1位点上,1、N和2*等3种亚基类型出现频率依次为88.10%、9.52%和 2.38%;在 Glu-B1 位点上,7、7+8、7+9、6+8 等 4 种亚基类型出现频率依次为 7.14%、40.48%、50%、2.38%;在Glu-D1位点上,2+12亚基类型出现频率为35.71%;优质亚基5+10出现的频率高达64.29%.亚基组合类型共有9种,1/7+9/5+10组合频率最高,为33.33%;1/7+8/5+10组合为23.81%;1/7+8/2+12组合为11.90%;其他6种组合频率均在10.00%以下.品质评分结果显示,自选品系得分为5~10分,平均得分8分,10个品系得分达10分.综合农艺性状鉴定,筛选出科大456、科大808和科洛5号等3个中强筋品种;科紫1号等19个品系的蛋白质含量达到强筋小麦标准.
High molecular weight wheat glutenin subunits(HMW-GS)are an important factor affect-ing wheat(Triticum aestivum L.)quality,and the analysis and selection of their composition is an important part of the research for the selection and breeding of new high-quality varieties.In this study,42 new wheat varieties(lines)were independently selected by the group as materials,and their HMW-GS composition and quality-related traits were identified based on comprehensive agronomic i-dentification,sodium dodecyl sulphate-polyacrylamide gel electrophoresis(SDS-PAGE),and molecu-lar markers.The results showed that a total of 9 subunit types of HMW-GS appeared in 42 new wheat lines.3 subunit types,1,N and 2*,were found at Glu-A1 locus with frequencies of 86.04%,11.63%and 2.33%,respectively;4 subunit types,7,7+8,7+9 and 6+8,appeared at Glu-B1 locus with frequencies of 6.98%,41.86%,48.84%and 2.33%;At Glu-D1 locus,the subunit type of 2+12 ap-peared with frequency was 35.71%;The frequency of high quality subunit 5+10 appeared as high as 64.29%.There were 9 types of subunit combinations,with the highest frequency of 33.33%for the 1/7+9/5+10 combination;23.81%for the 1/7+8/5+10 combination;11.90%for the 1/7+8/2+12 combination;and the frequencies of the other 6 combinations were all below 10%.The results of qual-ity scoring showed that the self-selected lines scored 5-10 points,with an average score of 8 points,and 10 lines scored up to 10 points.Comprehensive agronomic traits were identified to select and breed 3 medium-gluten varieties,KD 456,KD 808,and Kelo No.5.The protein content of 19 lines,inclu-ding Kezi No.1,reached the standard for high-gluten wheat.
