非靶向代谢组学分析不同山银花种质的化学成分差异OA
Untargeted Metabolomics Analysis of Chemical Composition Differences among Different Germplasms of Lonicerae Flos
为定向选育适宜贵州绥阳地区生态条件的高产优质山银花品种,采用超高效液相色谱-电喷雾电离-串联质谱(UPLC-ESI-MS/MS)与顶空-固相微萃取-气相色谱-质谱联用(HS-SPME-GC-MS)双平台非靶向代谢组学技术,对3份山银花(灰毡毛忍冬)种质(包括湘蕾金银花LF1及2份绥阳本地种质LF2、LF3)与1份忍冬种质"九丰1号"金银花(LF4)的花蕾进行代谢物检测与分析,系统比较不同忍冬种质花蕾的代谢物组成差异.主成分分析与正交偏最小二乘法分析(OPLS-DA)结果显示,不同种质间代谢物分离趋势显著.差异代谢物的KEGG通路富集分析表明,金银花LF4在黄酮类生物合成通路中代谢物含量较高(如木犀草素、槲皮素),而绥阳山银花LF2与LF3在单萜类生物合成通路中代谢物含量较高(如芳樟醇、薄荷醇).初步筛选出11种绥阳山银花与湘蕾金银花共有的差异代谢物,以及5种山银花与金银花共有的差异代谢物,这些代谢物可作为不同种质鉴别的潜在标志物.道地产区绥阳山银花种质(如LF2)在单萜类、酚酸类及皂苷类代谢物含量上具有显著优势,研究结果为山银花种质鉴别与良种选育提供了重要依据.
To conduct targeted breeding of high-yield and high-quality mountain Lonicerae Flos varie-ties suitable for the ecological conditions of Suiyang area in Guizhou Province,ultra-high performance liquid chromatography-electrospray ionization-tandem mass spectrometry(UPLC-ESI-MS/MS)and headspace-solid phase microextraction-gas chromatography-mass spectrometry(HS-SPME-GC-MS)dual-platform non-targeted metabolomics techniques were employed to detect and analyze the metabo-lites of 3 mountain Lonicerae Flos(Lonicera macranthoides)varieties(including Xianglei Lonicera japonica LF1 and 2 local germplasms from Suiyang LF2 and LF3)and 1 Lonicera japonica germ-plasm Jiufeng No.1(LF4).The differences in metabolite composition among the different Lonicerae germplasms were systematically compared.Principal component analysis and orthogonal partial least squares discriminant analysis(OPLS-DA)results showed that the separation trend of metabolites be-tween different varieties was significant.The KEGG pathway enrichment analysis of the differential metabolites indicated that the Lonicera japonica LF4 had higher content of metabolites in the flavonoid bio-synthesis pathway(such as luteolin and quercetin),while the local Lonicerae Flos LF2 and LF3 had higher content of metabolites in the monoterpenoid biosynthesis pathway(such as linalool and menthol).11 differen-tial metabolites common to both Suiyang Lonicerae Flos and Xianglei Lonicera japonica,and 5 differential metabolites common to both Lonicerae Flos and Lonicera japonica were preliminarily screened.These metab-olites could serve as potential markers for the identification of different varieties.The local Lonicerae Flos germplasm(such as LF2)from Suiyang had significant advantages in the content of monoterpenoids,phenolic acids,and saponins.The research results provided important basis for the identification of Lonicerae Flos germplasms and the selection of superior varieties.
