The regulatory role and research advances of STING inhibitors in post-stroke inflammation
Stroke is an acute neurological injury caused by a sudden interruption of cerebral blood supply, which imposes a heavy burden on patients due to its high mortality and disability rates.Following a stroke, the cGAS-STING pathway becomes activated, strongly inducing a type Ⅰ interferon response that ex-acerbates stroke-related damage.Targeted inhibition or degra-dation of STING can alleviate excessive inflammatory signaling and improve therapeutic outcomes.Strategies for STING inhibi-tion include the use of covalent inhibitors and competitive an-tagonists that target the ligand-binding domain(LBD)to block cyclic dinucleotide(CDN)binding, thereby suppressing STING activation. In addition, selective degradation of STING via the ubiquitin-proteasome system has also demonstrated promising efficacy. This review examines the role of the cGAS-STING pathway in stroke and summarizes recent advances in the development of STING inhibitors and degraders for the treat-ment of stroke. We aim to provide new perspectives and strate-gies to support the development of innovative anti-stroke thera-pies targeting STING.
