为了培育适应性强、抗逆性好的高羊茅新品种,利用RACE克隆技术从高羊茅干旱转录组中筛选并克隆了FabHLH63,进行生物信息学、亚细胞定位及干旱胁迫下表达分析.结果表明,获得FabHLH63基因完整cDNA序列1 311 bp,其编码一个含359个氨基酸残基的蛋白,具有典型的bHLH结构域;系统进化分析发现,与禾本科植物黑麦草(Lolium multiflorum)的bHLH蛋白亲缘关系较近;亚细胞定位显示FabHLH63蛋白定位于细胞核.实时荧光定量PCR结果表明,在干旱胁迫下,FabHLH63的表达显著上调,说明FabHLH63可能在抗旱响应中发挥了重要作用.研究结果为深入理解FabHLH63在高羊茅抗逆中的分子机制提供了理论依据,并为高羊茅分子育种提供了潜在的基因靶点.

In order to cultivate high-yielding and drought-resistant new varieties of Festuca elata,the RACE technology was used to screen and clone FabHLH63 from the drought transcriptome of Festu-ca elata.Bioinformatics,subcellular localization and expression analysis under drought stress were conducted.The results showed that the complete cDNA sequence of the FabHLH63 transcription fac-tor was 1 311 bp,encoding a protein containing 359 amino acid residues,with a typical bHLH do-main.The phylogenetic analysis revealed that it had a closer genetic relationship with the bHLH pro-tein of the grass species Lolium multiflorum.The subcellular localization indicated that the FabHLH63 protein was located in the nucleus.The results of real-time fluorescent quantitative PCR showed that the expression of FabHLH63 was significantly upregulated under drought stress,indica-ting that FabHLH63 could play an important role in the drought response.The research results pro-vided a theoretical basis for in-depth understanding of the molecular mechanism of FabHLH63 in the drought resistance of Festuca elata,and offerred potential gene targets for molecular breeding of Festuca elata.
