N-乙酰氨基葡萄糖转移酶V/α-1,6-甘露糖糖蛋白6-β-N-乙酰氨基葡萄糖转移酶(alpha-1,6-mannosylglycoprotein 6-beta-N-acetylglucosaminyltransferase,GnT-V/MGAT5)是调控N糖基化的重要酶类之一,负责合成β1,6位糖苷键的N-乙酰氨基葡萄糖胺(β1,6-GlcNAc)支链N-聚糖,参与调控肿瘤细胞的恶性表型.已有研究指出,GnT-V在肿瘤增殖、细胞黏附、侵袭迁移及免疫逃逸等方面扮演重要角色,但其在肿瘤中的详细作用机制尚未得到系统归纳和总结.因此,该文将全面探讨GnT-V如何介入肿瘤细胞恶性进展的调控过程,分析其在肿瘤糖生物学中的关键作用及潜在的临床应用前景,为靶向GnT-V的抗肿瘤药物开发提供理论依据.

GnT-V/MGAT5(alpha-1,6-mannosylglycoprotein 6-beta-N-acetylglucosaminyltransferase),a key enzyme regulat-ing N-glycosylation,is responsible for synthesizing β1,6-GlcNAc-branched N-glycans by catalyzing the formation of β1,6-glycosidic linkages.It plays a significant role in modulating the malignant phenotypes of tumor cells.Although previous studies have demonstrated the involvement of GnT-V in tumor proliferation,cell adhesion,invasion,migration,and immune evasion,its detailed mechanisms of action within tumors have not been systematically reviewed and summarized.Conse-quently,this article comprehensively explores how GnT-V par-ticipates in regulating malignant progression in tumor cells,ana-lyzes its pivotal role in tumor glycobiology,and discusses its po-tential clinical applications.This review aims to provide a theo-retical foundation for developing GnT-V-targeted anti-tumor therapeutics.
