菘蓝果实外果皮对自身种子萌发抑制作用及生理机制研究OA
Study on the Inhibitory Effect and Physiological Mechanisms of the Exocarp of Isatis indigotica Fruits on the Germination of Their Own Seeds
为探究菘蓝果实浸提液对其种子萌发的影响机制,根据菘蓝角果、种子的吸水特性以及生活力,确定菘蓝果实浸提液制备的提取时间,利用非靶代谢检测,筛选水溶性差异代谢物,并通过外源施加实验,分析该物质及原浸提液对菘蓝种子萌发早期(0~3 d)生理活性的影响.结果表明,菘蓝角果浸泡20 h达到种子吸胀状态,最大吸水率为291.0%,而菘蓝种子达到吸胀状态需要浸泡6 h,最大吸水率为60.5%;ck组发芽率为93.67%,角果浸提液组发芽率为6.67%,菘蓝种子浸提液组发芽率为69.33%;基于|log2(FC)|≥5筛选差异代谢物163种,其中水溶性差异代谢物41种.外施3-羟基-2-吡啶甲酸对菘蓝种子萌发具有较强的抑制作用;外施浸提液和3-羟基-2-吡啶甲酸溶液影响了菘蓝种子中淀粉、淀粉分支酶(SBE)、蔗糖、蔗糖酶、POD、可溶性蛋白、GA、IAA、ABA等含量和活性的变化.研究表明,菘蓝角果浸提液对自身种子萌发存在抑制作用,并且是多种物质共同作用,抑制物主要集中在菘蓝角果的外果皮部位,抑制物通过影响菘蓝种子的淀粉、可溶性蛋白、蔗糖酶、SBE及IAA等物质的代谢来调控种子萌发.
To explore the mechanism of the effect of the extract from the Isatis tinctoria fruit on the seed ger-mination,based on the water absorption characteristics and viability of the Isatis tinctoria siliques and seeds,the extraction time for the preparation of the Isatis tinctoria fruit extract was determined.Using non-target metabolite detection,water-soluble differential metabolites were screened,and through exogenous application experiments,the effects of this substance and the original extract on the physiological activity of Isatis tincto-ria seeds in the early stage(0-3 days)of germination were analyzed.The results showed that the Isatis tinc-toria siliques soaked for 20 hours reached the state of seed swelling,with the maximum water absorption rate of 291%,while the Isatis tinctoria seeds reached the state of swelling after soaking for 6 hours,with the maximum water absorption rate of 60.5%.The control group had a germination rate of 93.67%,the silique extract group had a germination rate of 6.67%,and the Isatis tinctoria seed extract group had a germination rate of 69.33%.163 differential metabolites with|log2(FC)|≥5 were screened,among which 41 were wa-ter-soluble differential metabolites.Exogenous application of 3-hydroxy-2-pyridinecarboxylic acid had a strong inhibitory effect on Isatis tinctoria seed germination.The exogenous application of the extract and 3-hydroxy-2-pyridinecarboxylic acid solution affected the changes in the contents and activities of starch,SBE,sucrose,sucrosease,POD,soluble protein,GA,IAA and ABA in Isatis tinctoria seeds.The study showed that the Isatis tinctoria silique extract had an inhibitory effect on its own seed germination,and it was the combined effect of multiple substances.The inhibitors were mainly concentrated in the exocarp of the Isatis tinctoria si-lique and the inhibitors regulated seed germination by affecting the metabolism of substances such as starch,soluble protein,sucrosease,SBE and IAA in Isatis tinctoria seeds.
