基于网络药理学与分子对接探讨温胆汤"异病同治"失眠和抑郁症的作用机制OA

Objective To explore the mechanism of Wendan Decoction for"treating different diseases with the same therapy"in the treatment of insomnia and depression using network pharmacology;To conduct verification through molecular docking.Methods The active components of Wendan Decoction were obtained through the TCMSP database and literature retrieval,and their action targets were predicted.The disease-related targets of insomnia and depression were retrieved in the GeneCards,OMIM and TTD databases,and the intersection with the action targets of the active components was taken.The STRING platform was used to construct the protein-protein interaction(PPI)network of the intersection targets,and NOC and Mcode were used to analyze the core targets.The Metascape platform was used to perform GO and KEGG pathway enrichment analysis on the intersection targets.Finally,AutoDock was used for molecular docking verification.Results Totally 60 active components and 321 targets of Wendan Decoction were screened.The core components in the formula included luteolin,quercetin,naringenin,β-carotene,baicalein,etc.The core intersection targets mainly included AKT1,IL6,TNF,IL1B,CYCS,etc.The KEGG pathway was mainly enriched in the pathways of IL-17,dopaminergic synapses,etc.The results of molecular docking showed that the binding activity between the core components and the core targets was good.Conclusion Wendan Decoction exerts the effect of"treating different diseases with the same therapy"in the treatment of insomnia and depression by inhibiting inflammatory reactions,promoting neuron repair and regulating neurotransmitters.
