肤痔清软膏调控铁死亡改善糖尿病足溃疡的机制研究OA
Mechanism of Fuzhiqing Ointment in Improving Diabetic Foot Ulcer by Regulating Ferroptosis
目的 本研究旨在明确肤痔清软膏对糖尿病足溃疡(DFU)小鼠模型的治疗作用,以及肤痔清软膏对溃疡模型中铁死亡活性的影响.方法 将 C57BL/6 雄性小鼠按随机区组法分为对照组、模型组、肤痔清软膏组(简称FZQ 组)和FZQ+铁死亡诱导剂厄拉司汀(Erastin)组(简称 FZQ+Erastin 组),除对照组外,其余组小鼠均给予链脲佐菌素并切除背部皮肤构建 DFU 模型,随后用肤痔清软膏涂抹于创面或联合腹腔注射 Erastin 进行干预;记录造模之后第 1、3、7、14 和 21 日的小鼠体质量、血糖和创面愈合情况;苏木精-伊红(HE)染色法检测皮肤组织病理变化并通过试剂盒检测皮肤组织中 Fe2+、丙二醛(MDA)和脂质过氧化物(LPO)的水平;蛋白质印迹法(Western blotting)检测皮肤组织中谷胱甘肽过氧化酶 4(GPX4)、溶质载体家族 7 成员 11(SLC7A11)和血红素加氧酶 1(HMOX1)的表达水平.结果 与对照组比较,模型组小鼠的体质量显著下降,而血糖水平明显增加,愈合效果也变得缓慢,同时皮肤组织存在纤维细胞排列紊乱,大量炎性细胞浸润的情况;在使用肤痔清软膏处理后,DFU 小鼠症状得到明显改善.在机制上,与对照组比较,模型组小鼠 MDA、LPO、Fe2+水平及 HMOX1 的表达显著增加,GPX4 和 SLC7A11 的表达受到抑制;肤痔清软膏可以抑制模型组小鼠铁死亡活性的升高,Erastin 可减弱肤痔清软膏对 DFU 的治疗效果,部分消除了肤痔清软膏对铁死亡活性的抑制作用.结论 肤痔清软膏可有效治疗 DFU 小鼠的症状,其发挥疗效的作用机制可能与调控铁死亡活性有关.
Objective In order to elucidate the therapeutic effect of Fuzhiqing ointment on mouse models of diabetic foot ulceration(DFU)and its impact on ferroptosis activity in ulcer models.Methods C57BL/6 male mice were randomly divided into control group,model group,Fuzhiqing ointment group(short for FZQ group)and FZQ+ferroptosis inducing agent(Erastin)group(short for FZQ+Erastin group)using a randomized block method.Except for the control group,mice in other groups were induced with diabetes by streptozotocin,and then underwent back skin excision to establish the DFU model.Then Fuzhiqing ointment was applied to the wound or combined with intraperitoneal injection of Erastin for intervention.The body weight,blood glucose and wound healing of mice were recorded on days 1,3,7,14 and 21 after modeling.The pathological changes of skin tissue were detected by hematoxylin-eosin(HE)staining,and the levels of Fe2+,malondialdehyde(MDA)and lipid peroxidation(LPO)in skin tissue were detected by kits.Western blotting was used to detect the expression levels of glutathione peroxidase 4(GPX4),solute carrier family 7 member 11(SLC7A11)and heme oxygenase 1(HMOX1)in skin tissue.Results Compared with the control group,the body weight of the mice in the model group decreased significantly,while the blood glucose level increased significantly,and the healing effect became slow.At the same time,disordered arrangement of fibroblasts and massive inflammatory cells infiltration were observed in the skin tissue of the model group.After Fuzhiqing ointment treatment,the symptoms of DFU mice were significantly improved.Mechanistically,compared with the control group,the levels of MDA,LPO,Fe2+and the expression of HMOX1 in the model group were significantly increased,while the expressions of GPX4 and SLC7A11 were inhibited.Fuzhiqing ointment inhibited the increasing of ferroptosis activity in the model group.However,Erastin treatment attenuated the therapeutic effect of Fuzhiqing ointment on DFU and partially reversed its inhibitory effect on ferroptosis.Conclusion Fuzhiqing ointment can effectively improve the symptoms of DFU in mice,and its therapeutic mechanism may be related to the regulation of ferroptosis activity.
