基于Hspa5/PrPc信号研究健脑解郁方对脑小血管病并发抑郁症大鼠海马和皮质星形胶质细胞铜稳态的影响OA
Exploration on the effects of Jiannao Jieyu Formula in improving copper homeostasis of cerebral small vessel disease related with depression rats based on Hspa5/PrPc signal
目的:探讨健脑解郁方对脑小血管病并发抑郁症(CSVDD)大鼠星形胶质细胞(AS)铜稳态的干预作用及机制.方法:40只自发性高血压大鼠随机分为模型组,阳性对照组,健脑解郁方低、中、高剂量组,每组8只,另取8只WKY大鼠作为对照组.大鼠背部皮下注射D-半乳糖8周联合双侧颈总动脉狭窄4周及慢性不可预见性温和刺激4周建立CSVDD模型.采用行为学实验评价大鼠抑郁样行为;LFB染色观察大鼠脑白质胼胝体病理变化;免疫组化技术检测海马、皮质AS的总突起长度和胶质纤维酸性蛋白阳性细胞数;ELISA和金属离子电子探针检测海马、皮质AS铜离子含量;免疫荧光检测AS中铜离子转运蛋白1(Ctr1)的表达;Western Blot、RT-qPCR技术检测海马、皮质中热休克蛋白家族A成员5(Hspa5)、朊蛋白(PrPc)的蛋白和mRNA表达.结果:与对照组比较,模型组大鼠自主活动减少,胼胝体髓鞘损伤,AS铜离子含量显著增加(P<0.01),Ctr1表达显著增加(P<0.01),Hspa5、PrPc蛋白和mRNA表达显著降低(P<0.01).与模型组比较,健脑解郁方高剂量组大鼠自主活动显著增加(P<0.01),摄食潜伏期显著降低(P<0.01);胼胝体空泡化改善,AS总突起长度显著增加(P<0.01,P<0.05),GFAP阳性细胞数显著减少(P<0.01),海马及皮质中铜离子含量显著降低(P<0.01),海马及皮质Ctr1表达显著降低(P<0.01),且海马和皮质中Hspa5、PrPc蛋白和mRNA表达水平显著增加(P<0.01,P<0.05).结论:健脑解郁方可能通过激活Hspa5/PrPc信号,改善铜稳态失衡,降低反应性AS增生,发挥缓解CSVDD大鼠抑郁样行为的作用.
Objective:To investigate the interventional mechanisms of Jiannao Jieyu Formula(JNJYF)on the copper homeostasis of astrocytes(AS)in rats with cerebral small vessel disease related with with depression(CSVDD).Methods:A total of 40 spontaneously hypertensive(SHR)rats were randomly divided into 5 groups:the model group,the positive control group,and the low,medium,and high dose groups of JNJYF,8 rats in each group,and 8 WKY rats were taken as the control group.D-galactose was injected subcutaneously into the back of rats for 8 weeks,together with bilateral common carotid artery stenosis for 4 weeks and chronic unpredicted mild stress for 4 weeks.The CSVDD model was established.The depression-like behavior of rats was evaluated by using the open field experiment and novel feeding experiment;LFB staining was used to observe the pathological changes in the corpus callosum;content of copper ions in hippocampus and cortex were detected by ELISA and electron microprobe for metal ions,immunohistochemical techniques were used to detect the total protrusion length and number of glial fibrillary acidic protein(GFAP)-positive cells in the hippocampal and cortex.Immunofluorescence were used to detect the expression of copper ion transporter protein 1(Ctrl),Western Blot and RT-qPCR techniques were used to detect the expression of heat shock protein family A member 5(Hspa5),prion protein(PrPc)in hippocampus and cortex.Results:Compared with the control group,rats in the model group exhibited reduced spontaneous activity,myelin damage in the corpus callosum,a significant increase in AS copper ion content(P<0.01),a significant increase in Ctr1 expression(P<0.01),and a significant decrease in Hspa5,PrPcprotein,and mRNA expression(P<0.01).Compared with the model group,rats in the high dose group of JNJYF exhibited significantly increased spontaneous activity(P<0.01)and significantly reduced feeding latency(P<0.01),corpus callosum vacuolization improved,total dendritic length in AS significantly increased(P<0.01,P<0.05),the number of GFAP-positive cells was significantly reduced(P<0.01),and copper ion level in the hippocampus and cortex was significantly decreased(P<0.01),in the Ctrl expression in the hippocampus and cortex was significantly reduced(P<0.01),and the protein and mRNA expression levels of Hspa5,PrPc in the hippocampus and cortex were significantly increased(P<0.01,P<0.05).Conclusion:JNJYF can effectively ameliorate depressive-like behaviors in CSVDD rats,the underlying mechanism may be closely associated with the regulation of Hspa5/PrPc signal,maitaining copper homeostasis and suppressing reactive AS.
