目的:探究羽调音乐对条件性恐惧小鼠恐惧记忆消退的作用机制.方法:28只C57BL6/J雄性小鼠随机分为空白组、恐惧组、暴露组和羽调组,每组7只.采用巴甫洛夫条件反射范式建立小鼠条件性恐惧模型,通过恐惧记忆保持实验(FCT)、高架十字迷宫实验(EPM)、旷场实验(OFT)分别检测小鼠的恐惧和焦虑状态,通过rs-fMRI观察小鼠大脑神经兴奋性和功能连接的情况.结果:行为学上,与空白组比较,恐惧组小鼠在FCT中的木僵时间和木僵时间百分率显著升高(P<0.01),在EPM中的进入开放臂时间(OT)值以及在OFT中的中心区运动时间(CWT)值均显著下降(P<0.01);与恐惧组比较,羽调组和暴露组小鼠在干预第14天和21天的木僵时间和木僵时间百分率均显著降低(P<0.01),暴露组优于羽调组(P<0.01);两组OT及CWT值均显著升高(P<0.01),且羽调组优于暴露组(P<0.01).小鼠fMRI分析结果显示,恐惧组小鼠双侧杏仁核、海马、视觉皮层、前额叶皮质、躯体感觉皮质等区域的ALFF显著升高(P<0.01);羽调组小鼠双侧杏仁核、海马、前额叶皮质和纹状体等区域的ALFF显著降低(P<0.01);暴露组小鼠左侧下丘脑、杏仁核、丘脑等区域的ALFF显著升高(P<0.01),右侧部分海马区的ALFF显著降低(P<0.01).功能连接分析发现,与空白组比较,恐惧组小鼠杏仁核与海马功能连接显著增加(P<0.01);羽调组小鼠杏仁核与海马功能连接显著降低(P<0.01);暴露组小鼠躯体感觉皮质与杏仁核功能连接减弱(P<0.01).结论:羽调音乐对促进恐惧记忆消退具有显著作用,其机制可能与调节大脑中与恐惧情绪相关的神经回路有关.

Objective:To investigate the mechanism of Yu tune music on fear memory extinction of conditioned fear mouse.Methods:Twenty-eight C57BL6/J male mice were randomly divided into control group,fear group,exposed group,and music group,7 in each group.A conditional fear model of mice was established using Pavlov conditioned reflex paradigm.The fear and anxiety states of mice were detected through fear conditioning test(FCT),elevated plus maze test(EPM),and Open field test(OFT),respectively.The excitability and functional connectivity of mouse brain neurons were observed through rs fMRI.Results:In terms of behavior,compared with the control group,freezing time(FT)value and FT percentage in FCT were significantly increased in the fear group(P<0.01),while ot value in EPM and CWT value in OFT were significantly decreased(P<0.01);Compared with the fear group,the FT value and FT percentage of mice in the music group and the exposed group were significantly lower on the 14th and 21st day of intervention(P<0.01),the exposed group was better than the music group(P<0.01),the OT and CWT values of the two groups were significantly higher(P<0.01),and the music group was better than the exposed group(P<0.01).The results of fMRI analysis of mice brain showed that ALFF in bilateral amygdala,hippocampus,visual cortex,prefrontal cortex,somatosensory cortex and other regions of mice in fear group increased significantly(P<0.01);The ALFF of bilateral amygdala,hippocampus,prefrontal cortex and striatum in music group was significantly decreased(P<0.01);ALFF in the left hypothalamus,amygdala,thalamus and other regions of the mice in the exposed group increased significantly(P<0.01),while Alff in the right part of the hippocampus decreased significantly(P<0.01).Functional connectivity analysis showed that compared with the control group,the functional connectivity between amygdala and hippocampus in the fear group was significantly increased(P<0.01);The functional connection between amygdala and hippocampus in music group was significantly decreased(P<0.01);The functional connection between somatosensory cortex and amygdala was weakened in the exposed group(P<0.01).Conclusion:Yu tune music significantly facilitates the extinction of fear memories,with its mechanism may be related to modulating neural circuits in the brain associated with fear emotions.
