目的:探究益气化浊汤改善2型糖尿病大鼠肝脏异位脂质沉积(ELD)及胰岛素抵抗(IR)疗效与机制.方法:高糖高脂诱导ZDF大鼠构建2型糖尿病模型,设正常组,模型组,高、低浓度益气化浊汤组及二甲双胍组,干预8周检测糖脂代谢、肝脂肪沉积,RNA-seq筛选差异lncRNA/mRNA,检测lnc-PID1/PID1及IRS-1/Akt/FoxO1通路表达;高糖高脂诱导BRL-3A细胞建立IR模型,经siRNA干扰、RIP-qPCR验证lnc-PID1与PID1结合及通路调控机制.结果:益气化浊汤显著降低2型糖尿病大鼠糖脂水平,改善IR,减轻肝损伤及脂肪沉积(P<0.05);该方可调控脂肪酸代谢通路,下调lnc-PID1/PID1,激活IRS-1/Akt,抑制FoxO1(P<0.05);细胞实验证实siRNA抑制lnc-PID1可增强该药对IR细胞的葡萄糖消耗,且lnc-PID1与PID1直接互作(P<0.05).结论:益气化浊汤通过抑制lnc-PID1/PID1,激活IRS-1/Akt通路,抑制FoxO1活性,改善肝IR与ELD.

Objective:To explore the efficacy and mechanism of Yiqi Huazhuo Decoction(YD)in ameliorating hepatic ectopic lipid deposition(ELD)and insulin resistance(IR)in type 2 diabetes mellitus(T2DM)rats.Methods:ZDF rats were fed a high-sugar and high-fat diet to establish a T2DM model,and divided into normal,model,low/high-dose YD and metformin groups.After 8 week intervention,glycolipid metabolism,hepatic fat deposition and gene/protein expression were detected,with differentially expressed lncRNA/mRNA screened by RNA-seq.IR model of BRL-3A cells was established,and the interaction between lnc-PID1 and PID1 was verified via siRNA interference and RIP-qPCR.Results:YD significantly reduced glycolipid levels,improved IR,alleviated liver injury and ELD(P<0.05),down-regulated lnc-PID1/PID1,activated IRS-1/Akt and inhibited FoxO1(P<0.05).Cell experiments suggested siRNA inhibiting lnc-PIDl enhanced IR cell glucose consumption and confirmed lnc-PID1-PID1 interaction(P<0.05).Conclusion:YD improves hepatic IR and ELD by inhibiting lnc-PID1/PID1,activating the IRS-1/Akt pathway and suppressing FoxO1 activity.
