目的:探讨益气活血通络方调控孤核受体4A(NR4A1)/Hippo/Yes激酶相关蛋白(YAP)途径抑制单侧输尿管梗阻(UUO)小鼠肾小管上皮细胞(TEC)G2/M期阻滞拮抗肾纤维化(RF)的机制.方法:40只雄性C57/BL6小鼠随机分为假手术组(Sham组)、模型组(UUO组)、益气活血通络方组(YQHX组)、NR4A1激动剂Csn-B组(Csn-B组),每组10只.除Sham组外,其余各组均进行UUO造模.HE、Masson和天狼星红染色观察肾脏病理变化,免疫组化法检测α-SMA和Vimentin的表达,Western Blot检测α-SMA、Vimentin、NR4A1及Hippo/YAP通路相关蛋白的表达,免疫荧光染色法检测YAP与TEAD1、Ki67与p-H3的共定位情况.体外采用HK-2细胞随机分为空白组(Con组)、TGF-β 1组(TGF组)、TGF-β 1+益气活血通络方组(TGF+YQHX组)、TGF-β1+Csn-B组,除Con组外其余各组均予以TGF-β 1刺激制备细胞纤维化模型,作用48 h,收集细胞.Western Blot检测α-SMA、Vimentin、NR4A1及Hippo/YAP通路相关蛋白的表达,免疫荧光染色法检测YAP与TEAD1、Ki67与p-H3的共定位情况.结果:体内实验:与UUO组比较,YQHX组和Csn-B组肾脏纤维化程度减轻,α-SMA和Vimentin表达显著降低(P<0.05),NR4A1蛋白表达、p-YAP/YAP、p-MST/MST和p-LATS/LATS蛋白表达比值显著升高(P<0.05),YAP与TEAD1、p-H3与Ki67的共定位情况减弱.体外实验:与TGF组比较,TGF+YQHX组及TGF+Csn-B组 α-SMA和Vimentin蛋白表达显著降低(P<0.05),NR4A1 蛋白表达及p-YAP/YAP、p-MST/MST、p-LATS/LATS蛋白表达比值显著升高(P<0.05);YAP与TEAD1、p-H3与Ki67的共定位情况减弱.结论:益气活血通络方可调控NR4A1/Hippo/YAP途径抑制TEC的G2/M期阻滞拮抗RF.

Objective:To investigate the mechanism of Yiqi Huoxue Tongluo Formula regulating the solitary nucleus receptor 4A(NR4A1)/Hippo/Yes kinase-associated protein(YAP)pathway to inhibit the G2/M phase arrest of renal tubular epithelial cells(TEC)in unilateral ureteral obstruction(UUO)mice against renal fibrosis(RF).Methods:A total of 40 male C57/BL6 mice were randomly divided into Sham group(Sham group),model group(UUO group),Yiqi Huoxue Tongluo Formula group(YQHX group),NR4A1 agonist Csn-B group(Csn-B group),with 10 mice in each group.UUO modeling was performed in all groups except sham operation group.HE,Masson,and Sirius red staining were used to observe the pathological changes in the kidney tissues.Immunohistochemistry was employed to detect the expression of α-SMA and Vimentin.Western Blot was used to detect the expression of α-SMA,Vimentin,NR4A1,and proteins related to the Hippo/YAP pathway.Immunofluorescence staining was utilized to detect the co-localization of YAP and TEAD1,as well as Ki67 and p-H3.In vitro,HK-2 cells were randomly divided into blank group(Con group),TGF-β1 group(TGF group),TGF-β1+YQHX group(TGF+YQHX group)and TGF-β1+Csn-B group.Except for the blank group,cells in the other groups were stimulated with TGF-β1 to establish a cell fibrosis model,which was treated for 48 hours,cells were collected.Western Blot was used to detect the expression of α-SMA,Vimentin,NR4A1 and proteins related to the Hippo/YAP pathway.Immunofluorescence staining was used to detect the co-localization of YAP and TEAD1,as well as Ki67 and p-H3.Results:In vivo experiments:compared with the UUO group,the degree of renal fibrosis was alleviated in the YQHX group and the Csn-B group,the expression of α-SMA and Vimentin was significantly decreased(P<0.05),while the expression of NR4A1 and the protein ratios of p-YAP/YAP,p-MST/MST and p-LATS/LATS was significantly increased(P<0.05),the co-localization of YAP and TEAD1,as well as p-H3 and Ki67,was weakened.In vitro experiments:compared with the TGF group,the expression of α-SMA and Vimentin was significantly decreased in the TGF+YQHX group and the TGF+Csn-B group(P<0.05),and the expression of NR4A1 and the protein ratios of p-YAP/YAP,p-MST/MST,and p-LATS/LATS were significantly increased(P<0.05),the co-localization of YAP and TEAD1,as well as p-H3 and Ki67,was weakened.Conclusion:Yiqi Huoxue Tongluo Formula can antagonize RF by regulating NR4A1/Hippo/YAP pathway to inhibit the G2/M phase arrest of TEC.
