猪卵母细胞体外成熟的影响因素及调控策略研究进展OA
Research Progress on the Influencing Factors and Regulatory Strategies of Porcine Oocytes Maturation in vitro
猪卵母细胞在生殖领域应用广泛,猪卵母细胞体外成熟(In Vitro Maturation,IVM)提高了该细胞的利用率,为相关研究与应用的推进提供了关键支撑.然而,IVM 的猪卵母细胞质量及其胚胎发育潜能仍然较低,限制了该技术的进一步应用.影响猪卵母细胞 IVM 的关键因素包括卵巢保存条件(温度及保存时间)、卵泡大小、卵母细胞采集方法、卵丘-卵母细胞复合体质量、培养条件、成熟培养时间、氧化应激和培养方法,这些因素均具有可调控性.针对性调控这些因素可有效促进猪卵母细胞 IVM,具体策略包括:规范操作步骤并优化培养条件,在IVM液中添加生长因子和卵母细胞分泌因子,添加抗氧化剂缓解卵母细胞氧化应激损伤,采用三维培养法和两步成熟法等新型培养方法.本文综述了影响猪卵母细胞 IVM 的因素及促进其 IVM 的调控策略,为提高猪卵母细胞 IVM 质量提供了参考.
Porcine oocytes have extensive applications in the field of reproduction.In vitro maturation(IVM)of porcine oocytes enhances their utilization rate,providing critical support for advancing related research and applications.However,the quality of IVM porcine oocytes and their embryonic developmental potential remain relatively low,limiting further application of this technology.Key factors influencing porcine oocytes IVM include ovarian preservation conditions(temperature and preservation time),follicle size,oocyte collection methods,quality of the cumulus-oocyte complexes,culture conditions,maturation culture time,oxidative stress and culture methods,all of which are adjustable.Targeted regulation of these factors can effectively promote IVM of porcine oocytes.Specific strategies include:standardizing operational procedures and optimizing culture conditions,adding growth factors and oocyte-secreted factors to the IVM medium,addition of antioxidants to mitigate oxidative stress damage in oocytes,and adopting novel culture methods such as three-dimensional culture method and two-step maturation method.This article reviews the factors influencing IVM of porcine oocytes and the regulatory strategies promoting IVM of porcine oocytes,which can provide reference for improving the quality of IVM of porcine oocytes.
