天名精内生真菌Alternaria arborescens 次生代谢产物及其抗肝癌作用初筛OA
Secondary metabolites of the endophytic fungus Alternaria arborescens from Tianmingjing(Herba Carpesii)and preliminary screening of their anti-hepatocellular carcinoma activities
目的 对天名精内生真菌的次生代谢产物及其抗肝癌活性进行研究,为寻找新的抗肝癌先导化合物提供参考.方法 采用现代分离技术对天名精活性菌株次生代谢产物进行分离纯化,运用现代波谱技术对分离得到的单体化合物进行结构鉴定;通过CCK-8 法测试单体化合物对HepG2 细胞的体外细胞毒活性,从而筛选天名精活性菌株的活性成分.结果 从天名精活性内生真菌Alternaria arborescens的大米发酵产物中共分离鉴定出 11 个单体化合物,包括 3 个大环化合物,即terpestacin(1)、10,11-dehydrocurvularin(2)、curvularin(3);5 个聚酮:stemphyperylenol(4)、altertoxin I(5)、altertoxin II(6)、alternariol(7)、al-ternariol-9-methyl ether(8);1 个倍半萜内酯,即telekin(9);1 个甾体类,即ergosterol peroxide(10);1 个单萜类化合物,即nec-triapyrone(11).化合物 1~10 均对HepG2 细胞有一定的抑制作用,其中化合物 1、2、7、9 显示出较强的抑制活性,IC50 值分别为(27.09±1.75)μM、(12.13±0.68)μM、(47.60±2.04)μM、(24.46±1.80)μM.结论 从天名精内生真菌Alternaria arborescens的大米发酵产物中分离鉴定出 11 个化合物,其中化合物 1~9、11 首次在该菌株中分离得到,验证了植物内共生理论,内生真菌可以产生与原植物相同或相似的活性分子,研究潜力巨大.
Objective To investigate the secondary metabolites of the endophytic fungus Alternaria arborescens from Tianmingjing(Herba Carpesii)and their anti-hepatocellular carcinoma activities,and to provide a reference for the discovery of novel lead compounds against hepatocellular carcinoma.Methods Secondary metabolites from an active fungal strain isolated from Tianmingjing(Herba Carpesii)were separated and purified using modern isolation techniques,and the structures of the isolated compounds were elucidated by spectroscopic methods.The in vitro cytotoxic activity of the isolated compounds against HepG2 cells was evaluated by the CCK-8 assay,so as to screen for active constituents from the active strain.Results A total of 11 compounds were isolated and identified from the rice fermentation products of the active endophytic fungus Alternaria arborescens from Tianmingjing(Herba Carpesii),including three macrocyclic compounds:terpestacin(1),10,11-dehydrocurvularin(2),and curvularin(3);five polyketides:stemphyperylenol(4),altertoxin I(5),altertoxin II(6),alternariol(7),and alternariol-9-methyl ether(8);one sesquiterpene lactone:telekin(9);one steroid:ergosterol peroxide(10);and one monoterpene compound:nectriapyrone(11).Compounds 1-10 showed inhibitory effects on HepG2 cells to varying degrees,among which compounds 1,2,7,and 9 exhibited relatively strong inhibitory activity,with IC50 values of(27.09±1.75)μM,(12.13±0.68)μM,(47.60±2.04)μM,and(24.46±1.80)μM,respectively.Conclusion Eleven compounds were isolated and identified from the rice fermentation products of the endophytic fungus Alternaria arborescens from Tianmingjing(Herba Carpesii).Among them,compounds 1-9 and 11 were isolated from this strain for the first time,which supports the plant endosymbiosis theory.These findings indicate that endophytic fungi can produce bioactive molecules identical or similar to those of the host plant and have substantial research potential.
