目的 运用高效液相色谱-四极杆飞行时间质谱(HPLC-Q-TOF-MS)技术鉴定八珍袋泡茶的化学成分,并结合网络药理学,探究八珍袋泡茶防治气血两虚证的潜在作用机制.方法 通过HPLC-Q-TOF-MS技术系统鉴定八珍袋泡茶的化学成分,结合口服生物利用度及类药五原则,筛选出八珍袋泡茶的潜在活性成分;利用在线靶点预测平台收集靶点与疾病的相关信息,构建核心靶点蛋白质-蛋白质相互作用(PPI)网络,并进行富集分析;应用DAVID生物信息学资源数据库富集关键靶点相关的主要生物通路,采用Cytoscape 3.7.1 软件构建八珍袋泡茶"药物-成分-靶点"互作网络图,进而分析八珍袋泡茶防治气血两虚证的作用机制.结果 共鉴定出 72 个化学成分,并在此基础上筛选出 50 个与防治气血两虚证相关的核心靶点.PPI网络分析结果显示,肿瘤蛋白P53(TP53)、表皮生长因子受体(EGFR)、蛋白激酶B(Akt1)、HSP90AA1 及CTNNB1 等靶标蛋白在网络拓扑图中与气血两虚证相关信号通路联系紧密.结合网络药理学结果还发现,八珍袋泡茶中的代表性活性成分,如阿魏酸、阿魏酸松柏苷、白术内酯II、3β-羟基苍术酮、鞣花酸、甘草苷和藁本内酯等,可能具有与调节气血两虚证相关蛋白关键位点结合的潜力.结论 初步鉴定了八珍袋泡茶的物质基础,并明确了该方可通过调控MAPK、JAK-STAT及PI3K-Akt等信号通路发挥防治气血两虚证的作用.

Objective To identify the chemical constituents of Bazhen tea bag by high-performance liquid chromatography-quadrupole time-of-flight mass spectrometry(HPLC-Q-TOF-MS)and,in combination with network pharmacology,to investigate the potential mechanism of Bazhen tea bag in the prevention and treatment of qi and blood deficiency pattern.Methods The chemical constituents of Bazhen tea bag were systematically identified by HPLC-Q-TOF-MS.Based on oral bioavailability and Lipinski's rule of five,the potential active constituents of Bazhen tea bag were screened.Targets and disease-related information were collected through online target prediction platforms,a protein-protein interaction(PPI)network for hub targets was constructed,and enrichment analysis was performed.Enrichment analysis was performed using the DAVID Bioinformatics Resources database to identify major biological pathways associated with key targets.Cytoscape 3.7.1 was used to construct a"drug-constituent-target"interaction network of Bazhen tea bag,and the mechanism of Bazhen tea bag in the prevention and treatment of qi and blood deficiency pattern was further analyzed.Results A total of 72 chemical constituents were identified,and on this basis,50 core targets related to the prevention and treatment of qi and blood deficiency pattern were screened.The PPI network analysis showed that target proteins such as tumor protein p53(TP53),epidermal growth factor receptor(EGFR),protein kinase B1(AKT1),HSP90AA1,and CTNNB1 were closely connected,in the network topology,to signaling pathways related to qi and blood deficiency pattern.Network pharmacology results further revealed that representative active constituents in Bazhen tea bag,such as ferulic acid,coniferin ferulate,atractylenolide II,3β-hydroxyatractylone,ellagic acid,liquiritin,and ligustilide,have potential for binding to key sites of proteins related to the regulation of qi and blood deficiency pattern.Conclusion The material basis of Bazhen tea bag was preliminarily identified,and it was clarified that this formula may exert preventive and therapeutic effects on qi and blood deficiency pattern through regulation of signaling pathways such as MAPK,JAK-STAT,and PI3K-Akt.
