不同生境烟草及周边植物病毒多样性及田间溯源分析OA

Analysis of virus diversity and field traceability of tobacco and surrounding plants in different habitats

[目的]明确云南省楚雄烟区不同生境烟草及周边植物的主要病毒种类和侵染类型,探明不同烟草病毒的田间毒源植物.[方法]利用RT-PCR技术对6个不同生境中烟草及周边植物的9种烟草常见病毒(TMV、CMV、PVY、TVBMV、ChiVMV、PCSV、INSV、TNSV和TSWV)进行检测、测序及系统进化分析.[结果]不同生境烟草及周边植物上检测到TMV、CMV、TVBMV、ChiVMV、PCSV和TSWV6种病毒,其中TMV为优势病毒,检出率为91.7%;TSWV次之,检出率为85.4%.烟草及周边植物复合侵染严重,复合侵染率高达97.9%,其中以TMV+TSWV+ChiVMV为优势侵染组合.6种烟草病毒病的田间毒源植物TMV为鱼眼草、青蒿、喜旱莲子草、鬼针草、甘蓝、黄瓜和玫瑰7种,ChiVMV为喜旱莲子草、青蒿、冬葵、辣椒、菜豆和水芹菜6种,TSWV为油菜、柑橘、木贼、鬼针草和甘蓝5种,TVBMV为鱼眼草、鬼针草、喜旱莲子草和甘蓝4种,CMV为喜旱莲子草和辣椒2种,PCSV为木贼和玫瑰2种.[结论]云南省楚雄烟区不同生境烟草及周边植物病毒病复合侵染现象严重,尤其以TMV、TSWV和ChiVMV 3种病毒检测率高、寄主范围广,应重点关注,在烟草种植期间烟田周边毒源植物的清除及传毒昆虫的防治可列为烟草病毒病的防治措施.

[Objective]The study aimed to identify primary virus species,infection types,and virus host plants of tobacco and surrounding plants in different habitats within Chuxiong tobacco-growing area,Yunnan province.[Method]By RT-PCR technique,nine common tobacco viruses(TMV,CMV,PVY,TVBMV,ChiVMV,PCSV,INSV,TZSV,TSWV)of tobacco and surrounding plants from six different habitats were detected,sequenced,and followed by phylogenetic analysis.[Result]Six types of viruses,namely TMV,CMV,TVBMV,ChiVMV,PCSV and TSWV,were detected in tobacco and surrounding plants from different habitats.Among them,TMV was the predominant virus,with a de-tection rate of 91.7%,followed by TSWV with a detection rate of 85.4%.The compound infection of tobacco and surrounding plants was se-vere,with a compound infection rate as high as 97.9%.The predominant infection combination was TMV+TSWV+ChiVMV.The field virus host plants for the six tobacco virus diseases were as follows,seven types including Dichrocephala auriculata,Artemisia carvifolia,Alter-nanthera philoxeroides,Bidens pilosa,Brassica oleracea,Cucumis sativus and Rosa rugosa for TMV;six types including A.philoxeroides,A.carv-ifolia,Malva verticillata,Capsicum annuum,Phaseolus vulgaris and Oenanthe javanica for ChiVMV;five types including Brassica napus,Citrus reticulata,Equisetum hyemale,B.pilosa and B.oleracea for TSWV;four types including D.auriculata,B.pilosa,A.philoxeroides and B.olera-cea for TVBMV;two types including A.philoxeroides and C.annuum for CMV;two types including E.hyemale and R.rugosa for PCSV.[Con-clusion]The compound infection of virus diseases in tobacco and surrounding plants from different habitats within Chuxiong tobacco-growing area of Yunnan province is severe.In particular,TMV,TSWV and ChiVMV have high detection rates and wide host ranges,which shall be given key attention.During the tobacco-planting period,the removal of virus host plants around tobacco fields and the prevention and control of virus-transmitting insects can be listed as preventive measures for tobacco virus diseases.
