湟水流域梨种质资源遗传多样性分析OA
Genetic diversity analysis of pear(Pyrus)germplasm resource in Huangshui Basin
[目的]明确青海地方梨种质资源的遗传多样性,促进地方梨种质资源的鉴定和合理利用提供理论基础.[方法]以湟水流域5个栽培区收集的29份梨种质资源为研究对象,对其22个表型性状(13个描述型性状和9个数量型性状)进行观测,结合SSR分子标记对资源进行遗传多样性分析.[结果]多样性指数分析表明,29份梨种质资源数量型性状的多样性(0.963)比描述型性状的多样性(0.915)更丰富.主成分分析表明,7个主成分因子的累计贡献率达78.812%,表型性状中花器官形态和叶片形状可作为资源鉴定的主要形态指标.11对SSR分子标记共检测出85个等位基因(Na),位点多态性信息指数(PIC)平均值为0.576,较好揭示资源的遗传多样性.20份青海地方品种平均香农指数(I)为0.840,多态性信息含量(PIC)为0.420,表明地方品种的遗传多样性较丰富.聚类分析结果表明,湟水流域梨种质资源主要有四类:秋子梨、白梨、新疆梨和西洋梨,除西洋梨外其他均属地方品种.栽培环境会影响品种的表型,地方品种中新疆梨与西洋梨遗传距离较近,秋子梨、白梨与西洋梨遗传距离较远.[结论]湟水流域梨种质资源,尤其是青海地方品种,遗传多样性丰富,具有开发价值,建议加强对青海地方梨属果树的鉴定、保护和开发利用.
[Objective]The study aimed to explore the genetic diversity of local pear species germplasm resources in Qinghai Huangshui Basin and provide a theoretical foundation for promoting the identification and rational and effective utilization of local pear germplasm resources.[Method]Twenty-nine pear germplasm samples collected from five cultivation areas in the Huangshui Basin were used as objects,and 22 phe-notypic traits(13 descriptive phenotypic traits and 9 quantitative characters)were observed.Combined with SSR molecular markers,the ge-netic diversity of pear species germplasm resources in the Huangshui Basin was analyzed.[Result]The diversity of quantitative characters(0.963)was richer than that of descriptive phenotypic traits(0.915)in germplasm resources through diversity index analysis.Principal component analysis indicated that the cumulative contribution rate of 7 principal component factors reached 78.812%,with the morphological characteristics of floral organs and leaf shape being the main identification morphological indicators in phenotypic traits.A total of 85 alleles were detected using 11 SSR molecular markers with an average polymorphic information content(PIC)of 0.576,which could effectively re-veal the genetic diversity of these resources.The average Shannon's index(I)of twenty Qinghai local varieties was 0.840 and the PIC was 0.420,indicating relatively rich genetic diversity among local varieties.Cluster analysis showed that the pear germplasm resources in the Huangshui Basin mainly included four categories:Pyrus ussuriensis,P.bretschneideri,P.sinkiangensis and P.communis,and the other three types were local native varieties except for P.communis.There was a certain relationship between the phenotypic differences among varieties and the cultivation environment.Among the local varieties,P.sinkiangensis had a closer genetic distance to P.communis,while P.ussuriensis and P.bretschneideri had a farther genetic distance from P.communis.[Conclusion]The pear germplasm resources in the Huangshui Basin,especially the local varieties of Qinghai,exhibit rich genetic diversity.It is recommended to strengthen the identification,conservation,and de-velopment of Qinghai local pear germplasm resources.
