烟草COPT基因家族鉴定及表达特征分析OA
Identification and expression analysis of tobacco COPT gene family
[目的]全面解析烟草COPT基因家族成员的特征、进化关系及表达调控模式,为后续筛选烟草抗逆品种提供理论依据.[方法]对烟草k326基因组范围内的COPT基因家族成员进行系统筛选,并对其理化性质、基因结构、保守基序、进化关系、共线性分析、蛋白互作等方面开展分析.探明COPT基因家族成员在不同组织及打顶处理、干旱条件和烟草疫霉侵染下的表达模式.[结果]在烟草k326基因组中,共定位到8 个COPT基因家族成员,它们非均匀地散布于7条染色体上,且均编码疏水性蛋白.Nt-COPT之间存在8 个共线性基因对且Ka/Ks值均小于1.基因上游启动子区域富含多种激素、生长发育、胁迫和光响应元件.转录组数据表明,大多数NtCOPT基因在茎部位高表达,具有一定的组织特异性;在打顶和干旱处理下,基因NtCOPT4、NtCOPT6和Nt-COPT8表达量均有提高;在烟草疫霉侵染后,大部分基因的表达量显著上调.[结论]8个NtCOPT基因家族成员在进化上相对保守,在不同组织、生物胁迫和非生物胁迫中表达模式不同,推测其在烟草生长发育、抗旱、抗烟草疫霉等方面发挥重要作用,该结果为进一步研究和利用烟草COPT基因奠定基础.
[Objective]The study aimed to comprehensively analyze the characteristics,evolutionary relationship and expression regulation mode of tobacco COPT gene family members,and provide theoretical basis for the subsequent screening of tobacco resistant varieties.[Meth-od]The study systematically identified the COPT gene family members in the tobacco k326 genome and analyzed its physicochemical proper-ties,gene structure,conserved motifs,evolutionary relationships,synteny and duplication analysis,and protein interactions.We also explored the expression patterns of COPT gene family members in different tissues such as roots,young leaves and shoot tips and under apical stress,drought conditions and tobacco blight infestation.[Result]A total of eight COPT genes were identified in the tobacco genome of k326 variety,unevenly distributed on seven chromosomes,all of which were hydrophobic proteins.Eight co-linear gene pairs existed among NtCOPT genes with Ka/Ks values less than 1,and the upstream promoter regions of the genes contained a variety of hormone-,growth-development-,stress-and light-responsive elements.The transcriptomic data indicated that most NtCOPT genes were highly expressed in the young leaf site with some tissue specificity.The expression of genes NtCOPT4,NtCOPT6 and NtCOPT8 was increased under apical and drought treatments;and the expression of most genes was significantly up-regulated after tobacco blight infestation.[Conclusion]Eight NtCOPT gene family members were relatively evolutionarily conserved and had different expression patterns in different tissues,biotic and abiotic stresses,which were hy-pothesized to play important roles in tobacco growth and development,drought resistance,and resistance to tobacco blight mold.The results laid the foundation for further research and utilization of tobacco COPT genes.
