石斛组培污染枝孢菌的鉴定及其敏感性杀菌剂筛选OA
Identification of Cladosporium spp.in contaminated Dendrobium tissue culture and screening of its sensitive fungicides
[目的]为了明确石斛组培中常见污染的真菌种类,通过室内测定抑菌效果筛选对其敏感的杀菌剂.[方法]采用直接菌落挑取法从石斛组培污染培养基上分离真菌,对其菌落和孢子进行形态特征观察;PCR扩增分离菌株的ITS、EF1-α和RPB2片段,通过测序和序列分析鉴定污染真菌种类,并对分离菌株进行室内杀真菌剂筛选.[结果]分离的SH-1和SH-2菌株形态与枝孢菌(Cladosporium spp.)相同,且2个菌株的ITS和EF1-α分别与粗糙枝孢菌(C.asperulatum)和贵州枝孢菌(C.guizhouense)的一致性以100%和99.59%为最高.进一步通过基于ITS和EF1-α序列的系统发育分析,将SH-1和SH-2分别鉴定为粗糙枝孢菌和贵州枝孢菌.所筛选的8种杀真菌剂中,SH-1菌株对多菌灵和噻菌灵的敏感性最高,而SH-2菌株只对多菌灵敏感;相反,腐霉利对两种枝孢菌的抑菌效果最差,浓度达到3 mg/L时仍无明显抑菌效果.进一步测定多菌灵对SH-1和SH-2的EC50分别为0.09 mg/L和0.11 mg/L,且0.25 mg/L的多菌灵均可完全抑制2个菌株在PDA上的生长.[结论]本研究明确了铁皮石斛组和卓花石斛组培污染的真菌分别为糙壁枝孢菌和贵州枝孢菌,8种杀真菌剂中多菌灵对2种枝孢菌的抑制效果最佳.研究结果为有效防控石斛组培生产中的真菌污染提供科学依据和实践指导.
[Objective]The present paper aimed to identify the common fungal species that contaminated the tissue cultures of Dendrobium spp.and to screen for sensitive fungicides by evaluating their inhibitory effects in vivo.[Method]Fungal strains were isolated using the direct colony picking method from Dendrobium spp.tissue cultures media,and the morphological characteristics of their colonies and spores were ob-served.The ITS,EF1-α and RPB2 fragments were amplified from genomic DNA of isolated strains via PCR.The species of contaminating fun-gi were identified through PCR products sequencing and their sequences analysis.Additionally,indoor screening of fungicides was conducted against the isolated strains.[Result]The isolated strains SH-1 and SH-2 exhibited the same morphology as Cladosporium spp.,and their ITS and EF1-α sequences showed the highest identities 100%and 99.59%to C.asperulatum and C.guizhouense,respectively.Further phyloge-netic analysis based on ITS and EF1-α sequences indicated that SH-1 and SH-2 were respectively identified as C.asperulatum and C.guizhouense.Among the eight screened fungicides,the strain SH-1 showed the highest sensitivity to carbendazim and thiabendazole,while strain SH-2 was only sensitive to carbendazim.In contrast,procymidone showed the weakest inhibitory effect against both Cladosporium spp.,with no significant inhibition observed even at a concentration of 3 mg/L.The EC50 values of carbendazim for SH-1 and SH-2 were further de-termined to be 0.09 mg/L and 0.11 mg/L,respectively.Moreover,0.25 mg/L of carbendazim could completely inhibit the growth of both strains on PDA.[Conclusion]The study identifies that the fungal contaminants in tissue cultures of the D.officinale and the D.anosmum are C.asperulatum and C.guizhouense,respectively.Among the eight tested fungicides,carbendazim demonstrates the most effective inhibition a-gainst the two Cladosporium spp..The findings provide a scientific basis and practical guidance for effectively preventing and controlling fun-gal contamination in Dendrobium spp.tissue cultures production.
