旨在从血睾屏障(blood-testis barrier,BTB)关键基因的表达差异、紧密连接蛋白ZO-1在生精细胞中的定位差异、miRNA与BTB关键基因的靶向调控关系的角度,探究BTB功能异常与种间杂交后代犏牛雄性不育的关系.本研究基于课题组前期的睾丸转录组数据进行基因集富集分析(GSEA)和BTB关键基因差异表达分析;采集健康的、性成熟的普通牛(n=3,2.5岁)、牦牛(n=3,4.5岁)、犏牛(n=3,4.5岁)的睾丸组织,利用蛋白质免疫印迹、免疫荧光染色技术检测紧密连接蛋白ZO-1在睾丸中的表达水平及细胞定位;对BTB相关基因的靶miRNA进行预测,并开展miRNA表达差异分析.GSEA富集分析结果显示,组织重塑、内皮屏障的建立、间隙连接等GO功能和KEGG通路显著富集且关键基因在犏牛睾丸组织中显著上调表达,其中与BTB形成相关的TJP1、TJP2、GJA1、F11R、TJAP1和CDH2基因的mRNA表达量在犏牛中显著高于牦牛和普通牛(P<0.05),ZO-1蛋白表达量在3种牛睾丸组织间差异不显著(P>0.05).免疫荧光显示牦牛和普通牛睾丸曲细精管中,ZO-1蛋白主要定位于精原细胞和精母细胞中间间隙,而ZO-1定位于犏牛精母细胞两侧间隙,其蛋白定位差异明显.生物信息学分析发现,bta-miR-191 与 TJP1 基因、bta-miR-93 和 bta-miR-218 与 GJA1 基因、bta-miR-200a 和 bta-miR-194 与 TJAP1 基因、bta-miR-218与CDH2基因存在潜在靶向调控关系,且这些miRNAs在犏牛睾丸中表达下调.犏牛睾丸组织中BTB关键蛋白ZO-1的细胞定位存在异常,且BTB相关基因表达可能受miRNAs影响.

This study aimed to investigate the relationship between abnormal blood-testis barrier(BTB)function and male sterility in interspecific hybrid offspring dzo(cattle-yak)from three perspectives:the expression differences of BTB key genes,the localization differences of tight junction protein ZO-1 in spermatogenic cells,and the targeted regulatory relationships between miRNAs and BTB key genes.Based on the previous testicular transcriptome data,gene set enrichment analysis(GSEA)and differential expression analysis of key BTB genes were performed.Testicular tissues of healthy and sexually mature cattle(n=3,2.5 years),yaks(n=3,4.5 years)and dzo(n=3,4.5 years)were collected.The expression and localization of tight junction protein ZO-1 in testis were detected by Western blot and immunofluorescence staining.Target miRNAs of BTB genes were predicted,and differential expression analysis of these miRNAs was performed based on previous miRNA-seq data.The GSEA enrichment analysis showed that GO functions and KEGG pathway genes related to tissue remodeling,establishment of endothelial barriers,and gap junctions were significantly enriched in the dzo testicular tissue.The relative mRNA expression levels of BTB formation-related genes,including TJP1,TJP2,GJA1,F11R,TJAP1,and CDH2,were significantly higher in dzo than in yaks and cattle(P<0.05).However,the relative protein expression level of ZO-1 exhibited no significant differences in testicular tissues among dzo,yak,cattle(P>0.05).Immunofluorescence results indicated that ZO-1 was mainly localized in the intercellular spaces between spermatogonia and primary spermatocytes in the seminiferous tubules of yaks and cattle;whereas in dzo,it was localized on both sides of the primary spermatocytes.The localization of ZO-1 showed obviously significant differences among the three bovine species.Bioinformatics analysis indicated that bta-miR-191 potentially targets TJP1,bta-miR-93 and bta-miR-218 potentially target GJA1,bta-miR-200a and bta-miR-194 potentially target TJAP1,and bta-miR-218 potentially targets CDH2.Moreover,these miRNAs were downregulated in the testes of dzo.The cellular localization of the key BTB protein ZO-1 exhibits abnormalities in dzo testicular tissues,and the expression of BTB-related genes may be regulated by miRNAs.
