旨在揭示陕北白绒山羊毛囊诱导阶段(E65)、器官形成阶段(E90)和细胞分化阶段(E120)差异表达的miR-497-5p在绒山羊毛乳头细胞中的分子调控机制,为提高绒山羊绒毛产量提供理论依据.本研究分析课题组前期绒山羊胚胎期皮肤组织miRNA-seq数据,统计miR-497-5p在E65、E90及E120时期差异表达情况;联合课题组前期RNA-seq数据,预测与毛囊发生发育相关的miR-497-5p靶基因;利用双荧光素酶报告系统验证miR-497-5p与潜在靶基因RSPO3的靶向关系;在毛乳头细胞中分别转染miR-497-5p mimics/mimics nc和inhibitor/inhibitor nc,利用RT-qPCR、WB及免疫荧光试验检测RSPO3在mRNA和蛋白水平的表达情况;进一步利用功能回补试验,验证miR-497-5p与RSPO3的靶向关系.通过联合分析筛选出252个miR-497-5p靶向的胚胎期差异基因,差异基因的GO富集分析结果显示,生长发育调控(regulation of developmental growth)等条目被显著富集;双荧光素酶试验结果表明miR-497-5p能够与RSPO3的3'UTR结合;RT-qPCR、WB及免疫荧光试验结果显示,过表达miR-497-5p后RSPO3在mRNA及蛋白水平的表达量显著降低(P<0.05),抑制miR-497-5p后其表达量显著升高(P<0.05);功能回补试验结果显示,与仅过表达RSPO3组相比,同时过表达miR-497-5p与RSPO3后细胞活力出现显著降低(P<0.01).本研究结果表明,miR-497-5p能够通过调控RSPO3影响毛乳头细胞增殖,揭示了 miR-497-5p调控绒山羊毛囊发生发育的分子机制.

This study aimed to reveal the molecular regulatory mechanism of differentially expressed miR-497-5p in dermal papilla cells during the induction(E65),organogenesis(E90),and cytodifferentiation(E120)stages of cashmere goats,providing a theoretical basis for improving cashmere yield.In this study,we analyzed the miRNA-seq data of embryonic skin tissues of cashmere goats and counted the differential expression of miR-497-5p in the periods of E65,E90,and E120.Potential target genes associated with hair follicle development were predicted using cross-analyzed with RNA-seq.The targeting relationship between miR-497-5p and the potential target gene RSPO3 was verified using a dual luciferase reporter system.The miR-497-5p mimics/mimics nc and inhibitor/inhibitor nc were transfected into dermal papilla cells.The expression of RSPO3 at the mRNA and protein levels was detected by RT-qPCR,WB and immunofluorescence,and the targeting relationship between miR-497-5p and RSPO3 was further verified using functional rescue assays.A total of 252 miR-497-5p-targeted embryonic differential genes were screened,and the results of GO enrichment analysis showed that terms such as regulation of developmental growth were significantly enriched.Dual luciferase assay showed that miR-497-5p could bind to the 3'UTR of RSPO3.RT-qPCR,WB and immunofluorescence showed that the overexpression of miR-497-5p resulted in a significant decrease in the expression of RSPO3 at the mRNA and protein levels(P<0.05),and RSPO3 expression was significantly increased after miR-497-5p inhibition(P<0.05).The results of functional rescue assay showed that cell viability was significantly reduced after simultaneous overexpression of miR-497-5p and RSPO3 compared with the RSPO3-only group(P<0.01).This study indicated that miR-497-5p regulated the proliferation of dermal papilla cells through targeting RSPO3,thereby revealing its molecular mechanism in hair follicle development of cashmere goat.
