一株砂生槐根际促生菌的鉴定及其生物学特性研究OA
Identification and biological characterization of a plant growth-promoting rhizobacterium isolated from Sophora moorcroftiana
砂生槐根际土壤中定殖着丰富的微生物,筛选出促生特性优异的菌株并研究其生长特性,可为特殊生境促生菌资源的开发利用提供理论基础.本研究采用土壤十倍稀释法从西藏地区生长的砂生槐根际土壤中分离获得一株促生性能优异的菌株zw27,经形态观察、生理生化特征测定及16S rRNA 基因序列分析对菌株zw27进行了鉴定.分别采用Salkowski比色法和钼锑抗比色法对菌株zw27产吲哚乙酸和溶磷能力进行了测定;通过单因素实验测定了菌株zw27对碳源、氮源和无机盐等营养物质的利用,以及对NaCl、酸碱度、干旱、温度的耐受性.研究结果表明,菌株zw27为解蛋白芽孢杆菌(Bacillus proteolyticus),菌株吲哚乙酸产量(43.468±2.818)μg·mL-1,溶磷量(65.580±3.509)μg·mL-1;菌株在以淀粉为碳源、酵母浸粉为氮源和氯化钾为无机盐的培养基中生长量最大;菌株在 NaCl浓度小于7%、pH为4~10、聚乙二醇浓度低于15%、温度为10~20℃的胁迫下,均有较好的耐受性.该研究可为砂生槐根际土壤中促生菌在高盐、干旱和高寒等条件下的开发利用提供菌种资源.
Many microorganisms were colonized in the rhizosphere soil of Sophora moorcroftiana.Screening and investigating the strains with excellent plant growth-promoting traits can provide a theoretical basis for the development and utilization of promoting bacterial resources of special habitat.In this study,a plant growth-promoting bacterial strain zw27 was isolated from the rhizosphere soil of S.moorcroftiana in Xizang region.The strain zw27 was isolated using the soil ten-fold dilution method.The strain zw27 was identified based on morphological observation,physiolgical and biochemical characterization,and 16S rRNA gene sequence analysis.The production of indole-3-acetic acid for strain zw27 was detected by Salkowski colorimetric method.The phosphate-solubilizing capacity of strain zw27 was assessed using molybdenum-antimony anti-colorimetric method.The single-factor experiment was used to study the types of nutrients and biological characteristics of the strain zw27.The results showed that the strain zw27 is Bacillus proteolyticus.The production of indole-3-acetic acid is(43.468±2.818)μg·mL-1.And phosphate-solubilizing activity is quantified as(65.580±3.509 μg)·mL-1.The strain zw27 achieved maximal growth when it is cultured separately with starch as the carbon source,yeast extract as the nitrogen source,and sodium chloride as the inorganic salt.The strain zw27 exhibits good tolerance under stress conditions including NaCl concentration below 7%,pH ranging from 4 to 10,polyethylene glycol concentration below 15%,and temperatures between 10℃and 20℃.This study can provide microbial strain resource for the development and utilization of plant growth-promoting bacteria in rhizosphere soil of S.moorcroftiana under conditions such as high salinity,drought,and low temperature.
