目的 建立超高效液相色谱-四极杆-飞行时间串联质谱法(UPLC-Q-TOF-MS/MS)分析安体威颗粒化学成分及入血成分.方法 采用ACQUITY UPLC HSS T3(2.1 mm×100 mm,1.7 μm)色谱柱和0.1%甲酸(A)-乙腈(B)流动相系统进行梯度洗脱;体积流量0.3 mL/min;温度30℃;电喷雾离子源,正负离子模式扫描;根据准分子离子、保留时间、二级碎片离子结合对照品信息及自建数据库进行成分鉴定.结果 分析鉴定出55个体外化学成分.从大鼠血清中分析鉴定出37个成分,其中有22个原型成分,15个代谢产物.结论 建立了快速且稳定的安体威颗粒体内外化学成分分析方法,为安体威颗粒药效物质基础的阐明和质量标准的提升奠定基础.

Objective To establish a method based on ultra-high performance liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry(UPLC-Q-TOF-MS/MS)for analyzing the chemical constituents and blood-absorbed constituents of Antiwei Granule(安体威颗粒,ATWG).Methods The analysis was performed on an ACQUITY UPLC HSS T3 column(2.1 mm×100 mm,1.7 μm)maintained at 30℃.The mobile phase consisted of 0.1%formic acid in water(A)and acetonitrile(B)with a flow rate of 0.3 mL/min in a gradient elution mode.An electrospray ionization(ESI)source was used for detection in both positive and negative ion scanning modes.Constituents were identified based on their exact molecular ions,retention times,and MS/MS fragment ions,with reference to standard substance information and a self-built database.Results A total of 55 chemical constituents were identified from the granules in vitro.From rat serum samples,37 constituents were detected,of which 22 were prototype constituents and 15 were metabolites.Conclusion A rapid and reliable analytical method was established for identifying the chemical constituents of ATWG both in vitro and in vitro.This method lays a foundation for clarifying the pharmacodynamic material basis and improving the quality stan-dards of ATWG.
