目的 研究精天颗粒拆方对小鼠慢性肾纤维化的作用及其作用机制.方法 C57BL/6小鼠随机分为空白组、模型组、大黄甘草汤组、五苓散组、黄精红景天穿山龙组、精天颗粒组及阳性药组,每组8只.除空白组外各组使用0.2%腺嘌呤饲料喂养4周建立慢性肾纤维化模型,除空白组及模型组外各组分别同步予以相应药物治疗2周.观察小鼠一般体征;检测血清中肌酐及尿素水平;苏木素-伊红(HE)和高碘酸-席夫(PAS)染色检测肾组织病理变化;Masson(Masson trichrome stain-ing)染色观察肾脏纤维病理变化;Western blot(WB)检测各组小鼠肾组织中Wnt3a和β-连环蛋白含量(β-catenin)、α-平滑肌肌动蛋白(α-SMA)、Ⅰ型胶原蛋白(Col Ⅰ)、Ⅲ型胶原蛋白(Col Ⅲ)表达水平.结果 精天颗粒及其拆方明显改善肾纤维化小鼠体征,降低慢性肾纤维化小鼠血清中尿素及肌酐水平(P<0.01,P<0.01),改善肾脏组织病理变化,下调Wnt3a、β-catenin、Col Ⅰ、Col Ⅲ、α-SMA蛋白表达(P<0.05).全方组综合疗效最优,拆方组在不同指标上各具优势.结论 精天颗粒通过对Wnt/β-catenin通路的调控能有效抑制细胞外基质(ECM)过度沉积,改善慢性肾纤维化.

Objective To study the effect of Jingtian Granule(精天颗粒,JTG)disassembled formula on chronic renal fibrosis in mice and its mechanism of action.Methods C57BL/6 mice were randomly divided into the blank group,model group,Dahuang Gancao Decoction(大黄甘草汤,DHGCD)group,Wuling Powder(五苓散,WLP)group,Huangjing Hongjingtian Chuangshanlong(黄精红景天穿山龙,HHC)group,JTG group and positive drug group,with 8 mice in each group.In addition to the blank group,each group was fed with 0.2%adenine feed for 4 weeks to establish the chronic renal fibrosis model,and from the 3rd week onwards,each group was treated with the corresponding drugs for 2 weeks in addition to the blank group and the model group.General signs of mice were observed;creatinine and urea levels in serum were detected;pathological changes in renal tissues were detected by hematoxylin-eosin(HE)and periodate-schiff(PAS)staining;pathological changes in renal fibers were observed by Masson trichrome staining;and Wnt3 in renal tissues of mice in each group was detected by Western blot(WB).WB was used to detect the expression levels of Wnt3a and β-catenin,α-smooth muscle actin(α-SMA),collagen type I(ColⅠ)and collagen type Ⅲ(ColⅢ)in the kidney tissues of mice.Results JTG and its disassembled formula significantly improved the signs of mice with renal fibrosis,reduced the levels of urea and cre-atinine in the serum of mice with chronic renal fibrosis(P<0.01,P<0.01),improved the histopathological changes of kidneys,and down-regulated the protein expression of Wnt3a,β-catenin,ColⅠ,ColⅢ,α-SMA(P<0.05).The comprehensive efficacy of the whole formula group was optimal,and the disassembled formula group had its own advantages in different indexes.Conclusion JTG dis-assembled formula can effectively inhibit the excessive deposition of extracellular matrix(ECM)and alleviate chronic renal fibrosis through the regulation of Wnt/β-catenin pathway.
