基于机器学习与WGCNA综合分析CD5L在慢加急性肝衰竭中的免疫调节作用OA
The Immunomodulatory Role of CD5L in Slow-on-Chronic Acute Liver Failure Based on Machine Learning and WGCNA:a Comprehensive Analysis
目的 探讨 CD5L 在慢加急性肝衰竭(ACLF)中的免疫调节作用及其机制研究.方法 基于 GEO 数据库GSE139602 数据集,结合差异表达分析、GO 与 KEGG 通路富集、免疫浸润分析、WGCNA 及 SVM-RFE 算法,筛选 ACLF 关键基因,并通过组织病理染色与 qPCR 验证 CD5L 的表达水平.结果 共识别 1080 个差异表达基因,富集于胶原外基质-受体相互作用、胆汁分泌、叶酸代谢等通路.ACLF 组中性粒细胞显著上调(P<0.05).综合分析确定 CD5L 为关键调控基因,实验验证其在 ACLF 肝组织中显著下调(P<0.05),且与肝小叶结构损伤呈负相关.结论 CD5L 可能通过调控免疫微环境失衡促进 ACLF 进展,是潜在的诊断标志物和干预靶点.
Objective To investigate the immunomodulatory role of CD5L in acute-on-chronic liver failure(ACLF)and explore its mechanism of action.Methods Gene expression data from the GSE139602 dataset(GEO)were analyzed via differen-tial expression analysis,GO/KEGG enrichment,immune infiltration profiling,WGCNA,and SVM-RFE to identify key genes in ACLF.CD5L expression was validated via histopathology and qPCR.Results Total 1,080 differentially expressed genes(DEGs)were identified,enriched in pathways such as ECM-receptor interaction,bile secretion,folate metabolism,and cytochrome P450-me-diated detoxification.Neutrophil infiltration levels were significantly higher in ACLF liver tissues(P<0.05).CD5L was identified as a hub gene,and its expression was significantly downregulated in ACLF tissues(P<0.05),negatively correlating with lobular structural damage.Conclusion CD5L may contribute to ACLF progression by disrupting immune homeostasis and hepatic meta-bolic balance.It represents a potential diagnostic biomarker and therapeutic target.
