柳叶沙棘黄酮的绿色提取及抗氧化活性分析OA

目的 对柳叶沙棘黄酮成分进行绿色提取,鉴定其黄酮成分及抗氧化活性分析.方法 采用超声波辅助低共熔溶剂,筛选最佳提取溶剂并观察样品微观变化,通过单因素及响应面法优化提取工艺,分析黄酮成分及抗氧化能力.结果 确定氯化胆碱-乳酸为最佳提取溶剂,响应面优化得到最佳提取工艺参数为液料比 40∶1(mL∶g)、提取温度 55℃和提取时间 33 min,在此条件下黄酮得率为(77.12±0.12)mg·g-1.高效液相色谱鉴定出芦丁、异槲皮苷、槲皮苷以及槲皮素4 种黄酮成分.对2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS+)和一氧化氮清除剂(PTIO)抗氧化清除能力的 IC50值分别为 0.39 mg·mL-1和 1.17 mg·mL-1.结论 超声波辅助低共熔溶剂提取柳叶沙棘黄酮具有良好的应用前景.

Objective To achieve green extraction of flavonoids from Hippophae rhamnoides,identify the flavonoid components,and analyze their antioxidant activity.Methods An ultrasound-assisted deep eutectic solvent(DES)system was employed.The optimal DES was screened,and microstructural changes in the samples were observed.The extraction process was optimized through single-factor experiments and response surface methodology.The flavonoid composition was analyzed,and antioxidant capacity was evaluated.Results Choline chloride—lactic acid was identified as the optimal DES.The optimized extraction parameters were a liquid-solid ratio of 40∶1(mL∶g),a temperature of 55℃,and a time of 33 min,yielding a flavonoid content of(77.12±0.12)mg·g-1.High-performance liquid chromatography identified four flavonoids:ru-tin,isoquercitrin,quercitrin,and quercetin.The extract exhibited antioxidant activity with IC50 values for the antioxidant scav-enging capacity against ABTS+and PTIO were 0.39 mg·mL-1 and 1.17 mg·mL-1,respectively.Conclusion The ultrasound-assisted deep eutectic solvent method shows promising potential for the extraction of flavonoids from Hippophae rhamnoides.
