浅表局麻镇痛的中药面膜的制备及作用机制初探OA

Objective To develop an analgesic facial mask based on Chinese herbal extracts for local anesthesia before superficial minimally invasive medical art,and evaluate its analgesic effect and mechanism,in order to solve the clinical problems of cumbersome operation and uneven analgesia before superficial medical art,such as Shuiguang needle.Methods Using Ephedra,Asarum,Aconite and Glycyrrhiza as raw materials,the water extract and alcohol extract were prepared re-spectively and the traditional Chinese medicine facial mask was made.The rat self-control test(n=6)was used to divide the lumbar back skin into four areas,and the negative control(normal saline),positive control(lidocaine),compound water ex-tract and alcohol extract facial mask were applied randomly for 30 min,and the pain degree was comprehensively evaluated by acupuncture.Evaluate skin safety through H&E staining.Construct HEK-293T cell models overexpressing TRPA1 chan-nels and investigate the effects of each group on TRPA1 mediated calcium influx.Results The superficial analgesic effect of compound alcohol extract facial mask was equivalent to that of lidocaine,while the analgesic effect of water extract was weaker than that of alcohol extract;The skin structure of each group is intact,without inflammation or tissue damage;Both the water extract and alcohol extract of the compound can activate TRPA1 channels,and the activation intensity of the alco-hol extract is similar to that of the positive control.Conclusion Alcohol extract facial mask of Ephedra Asarum Aconite Gly-cyrrhiza has the similar analgesic effect of superficial local anesthesia to lidocaine,and its mechanism may be related to acti-vating TRPA1 channel and inducing its desensitization.
