茯苓水提物对抗柴胡皂苷 A 引起的肝损伤机制OA

目的 研究茯苓水提物缓解柴胡皂苷 A(SSA)所致肝损伤的作用和机制.方法 将 C57BL/6 小鼠随机分为正常对照组、SSA 模型组、茯苓低剂量治疗组和茯苓高剂量治疗组,通过灌胃 SSA 建立肝损伤模型,灌胃茯苓水提物予以治疗.检测小鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST)水平,及血清和肝脏中甘油三酯(TG)、胆固醇(TC)水平.采用网络药理学方法筛选茯苓作用的 SSA 的毒性靶点,从中构建细胞死亡相关通路.体外实验使用 AML12 肝细胞,检测茯苓水提物和 SSA 对细胞存活率的影响,并通过免疫印迹检测细胞死亡相关蛋白表达.结果 茯苓高剂量组可提升小鼠生存率,降低小鼠血清 ALT 和 AST 水平(P<0.001,P<0.01)以及血清与肝脏 TC、TG水平(P<0.05).网络药理学筛选获得 46 个茯苓干预 SSA 肝毒性的潜在靶点.KEGG 分析提示凋亡信号通路等可能参与调控,关键靶点包括 Bcl-2、CASP3、CASP9 等.细胞实验显示,茯苓高剂量组可提高细胞存活率(P<0.05),降低 cleaved-caspase-3、cleaved-caspase-9 蛋白表达(P<0.05,P<0.01),增强 Bcl-2 蛋白表达(P<0.01).结论 茯苓水提物对 SSA 诱导的肝损伤具有改善作用,其机制可能与阻断 SSA 引发的线粒体介导的凋亡通路有关.

Objective To study the effect and mechanism of water extract of Poria cocos in alleviating liver injury caused by saikosaponin A(SSA).Methods C57BL/6 mice were randomly divided into normal control group,saikosaponin A model group,low-dose Poria cocos treat group and high-dose Poria cocos treat group.A liver injury model was established by intragastric administration of SSA,the treatment was given by gavage of the water extract of Poria cocos.Detect the levels of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in the serum of mice,as well as the levels of triglycerides(TG)and cholesterol(TC)in the serum and liver.The toxic targets of SSA acting on Poria cocos were screened by network pharmacology methods,and the cell death-related pathways were constructed from them.In vitro experiments were conducted using AML12 hepatocytes to detect the effects of water extract of Poria cocos and SSA on cell survival rate.The effects of the water extract of Poria Cocos and SSA on cell survival rate were detected,and the expression of cell death related proteins was detected by Western blotting.Results The high-dose Poria cocos group improved survival rates in mice,significantly reduced the levels of serum ALT and AST in mice(P<0.001,P<0.01),and the contents of TC and TG in serum and liver(P<0.05).Network pharmacology screening identified 46 potential targets for Poria cocos intervention in SSA-induced hepatotoxicity.KEGG analysis suggested involvement of the apoptosis signaling pathway,with key targets including Bcl-2,CASP3,and CASP9.In cell experiments demonstrated that the high-dose Poria cocos group could significantly increase cell survival rate(P<0.05),down-regulate the protein expressions of cleaved-caspase-3 and cleaved-caspase-9(P<0.05,P<0.01),and enhance the protein expression of Bcl-2(P<0.01).Conclusion Poria cocos water extract significantly ameliorates SSA-induced liver injury,the mechanism may be related to blocking the mitochondrial-mediated apoptotic pathway triggered by SSA.
