目的 比较小鼠不同来源的骨源性间充质干细胞的成骨能力,为骨组织工程学研究提供成骨高效诱导技术体系.方法 将BALB/c小鼠腿骨骨髓(bone marrow,BM)、皮质骨片消化(bone digestion,BD)和皮质骨片培养(bone culture,BC)获得的间充质干细胞(mesenchymal stem cells,MSCs)通过传代和CCK8实验比较其增殖能力;通过流式细胞仪检测其干细胞占比情况.经成骨诱导后,通过碱性磷酸酶和茜素红染色、Western印迹法、实时定量PCR实验检测其体外成骨能力,明确最佳细胞类型.将该细胞在β-TCP上进行3D诱导培养2周后,通过Western印迹法、实时定量PCR实验检测其成骨能力.将3种细胞移植入BALB/c小鼠背部皮下,8周后取出样本,通过组织学染色检测成骨情况.结果 MSC-BD和MSC-BC较MSC-BM增殖能力更强;MSC-BC较MSC-BD在Runx2、Sp7等mRNA和蛋白表达水平明显上调,碱性磷酸酶与茜素红染色强度也明显增强;MSC-BM和MSC-BD组HE染色仅见纤维结缔组织形成,MSC-BC组HE染色见新骨形成.免疫组织化学染色结果显示MSC-BC组相对于MSC-BD组有更多OCN阳性细胞,而在MSC-BM中没有明显阳性细胞.结论 MSC-BC具有更强的增殖能力、更高的干细胞占比和成骨能力.

Objective To compare the osteogenic potential in mouse bone derived mesenchymal stem cells from different sources and provide an efficient osteogenic induction technical system for bone tissue engineering research.Methods Bone-derived mesenchymal stem cells were isolated from the leg bones of BALB/c mice through three sources:bone marrow(bone marrow,BM),cortical bone fragments digestion(bone digestion,BD),and cortical bone fragment culture(bone culture,BC).The proliferative capacity of the cells were compared via passaging and cell counting kit-8(CCK-8)assays,while flow cytometry was used to assess the stem cell proportions.Following osteogenic induction,osteogenic potential was evaluated using alkaline phosphatase(ALP)and Alizarin red staining,Western blotting,and real-time quantitative PCR,and the optimal cell type was determined.After inducing the selected cells in 3D culture on β-tricalcium phosphate(β-TCP)for 2 weeks,their osteogenic potential was assessed through Western blotting and real-time quantitative PCR experiments.The three types of cells were subcutaneously transplanted into the dorsal region of BALB/c mice.The samples were collected after 8 weeks,the osteogenesis was evaluated with histological staining.Results The MSC-BD and MSC-BC exhibited stronger proliferative capacity than MSC-BM.Compared with MSC-BD,MSC-BC showed significantly upregulated mRNA and protein expression levels of osteogenic markers(Runx2,Sp7),as well as enhanced ALP and Alizarin Red staining intensity.The HE staining of graft sections from MSC-BM and MSC-BD showed only fibrous connective tissue formation,while new bone formation was found with MSC-BC.The immunohistochemical staining results showed that MSC-BC had more osteocalcin-positive cells when compared with that in MSC-BD,while no significant positive cells were observed in MSC-BM.Conclusion The MSC-BC demonstrates superior proliferative capacity,higher stem cell proportions,and stronger osteogenic potential.
