抑郁症慢性不可预知温和刺激动物模型分析及评价OA
Analysis and Evaluation of Chronic Unpredictable Mild Stimulus Animal Model for Depression
目的 分析国内抑郁症慢性不可预知温和刺激(chronic unpredictable mild stimulation,CUMS)动物模型的造模特点,探索更符合疾病的抑郁症模型.方法 以"CUMS"和"动物模型"为主题词,检索中国知网数据库建库至2024年5月动物实验的相关文献,归纳动物种类、造模方式、检测指标等.SD雄性大鼠适应性喂养1周后随机均分为对照组和CUMS模型组,两组按照不同的造模方式处理,21 d造模结束进行相关行为学检测及海马HE染色.结果 共纳入标准文献332篇,动物多为雄性,以SD大鼠(226次,68.07%)、Wistar大鼠(40次,12.05%)、C57BL/6J小鼠(30次,9.04%)居多;造模周期以21 d(75次)、42 d(67次)和28 d(65次)3种为主.造模成功与否主要依据动物体质量、行为学、分子检测等方面,病理形态学观察主要采用海马组织HE染色.CUMS联合孤养造模21 d,模型组与对照组相比,体质量、进食量、蔗糖偏好、旷场总距离均下降,强迫游泳漂浮次数和时间均增加(P<0.05),模型组海马HE染色显示神经元排列疏松,结构欠清晰.结论 CUMS联合孤养模型体质量、进食量、蔗糖偏好、旷场总距离下降,静止时间增长,造模基本成功.
Objective To analyze the modeling characteristics of chronic unpredictable mild stimulation(CUMS)animal models of depression in China,and explore depression models that are more suitable for the disease.Methods Using"CUMS"and"animal models"as keywords,search for relevant literature on animal experiments from the China National Knowledge Infrastructure database until May 2024,and summarize animal species,modeling methods,detection indicators,etc.After one week of adaptive feeding,male SD rats were randomly divided into a control group and a CUMS model group.The two groups were treated according to different modeling methods,and after 21 days of modeling,relevant behavioral tests and hippocampal HE staining were performed.Results A total of 332 standard literature were included,with male animals being the majority,including SD rats(226 times,68.07%),Wistar rats(40 times,12.05%),and C57BL/6J mice(30 times,9.04%);The main molding cycles are 21 days(75 times),42 days(67 times),and 28 days(65 times).The success of modeling is mainly based on animal body mass,behavior,molecular testing,etc.The pathology was mostly hippocampal HE staining.After 21 days of CUMS combined with solitary confinement,the model group showed decrease in body weight,food intake,sucrose preference,and total distance of open field compared to the control group.The number and duration of forced swimming and floating increased(P<0.05).Hippocampal tissues were subjected to hematoxylin and eosin(HE)staining for pathological morphological evaluation.Conclusion The CUMS combined with solitary care model showed a decrease in body weight,food intake,sucrose preference,and total open field distance,with an increase in resting time,and the modeling was basically successful.
