肉桂维管组织发育的形态学研究OA
Morphological study on Cinnamomum cassia vascular tissue development
[目的]旨在揭示肉桂维管组织在发育过程中细胞形态、结构以及细胞壁主要成分木质素和纤维素的沉积规律.[方法]采集自茎尖至半木质化阶段的茎段,作为关键发育期的代表性样本.样本经石蜡包埋切片后,采用番红-固绿染色进行组织学观察,利用光学显微镜系统分析维管组织发育过程中的细胞形态与结构变化.使用Wiesner反应与自发荧光相结合的方法,特异性检测木质素的沉积,并采用钙荧光白对纤维素进行荧光标记与分析.[结果]肉桂维管组织的发育始于芽顶端分生组织细胞,逐渐分化为原形成层原始细胞;原形成层原始细胞进一步发育成原形成层细胞和韧皮纤维细胞.原形成层细胞一部分分化为初生韧皮部和初生木质部,另外一部分逐渐发育为形成层;形成层细胞将进一步分化为次生韧皮部和次生木质部.在次生生长阶段,可以观察到韧皮射线细胞存在活跃的平周分裂,可能参与了次生韧皮部细胞的形成.随着维管组织的发育,韧皮纤维细胞壁增厚,木质素和纤维素含量均较高;木质部始终维持较高的木质素含量,形成层细胞几乎不含木质素,纤维素含量较木质部和韧皮部低.[结论]研究揭示了肉桂初生维管组织和次生维管组织的发育轨迹,以及发育过程中木质素和纤维素在细胞壁沉积的动态变化和特点,为进一步深入研究肉桂维管组织发育的调控机制奠定基础,也为中药材肉桂品质形成机制的研究提供了有价值的线索.
[Objective]This study aims to elucidate the morphological and structural changes of vascular tissues in C.cassia during development,as well as the deposition patterns of the major cell wall components,lignin and cellulose.[Method]Stem segments ranging from the shoot apex to the semi-lignified stage were collected as representative samples of key developmental stages.After paraffin embedding and sectioning,the samples were stained with safranin-O and fast green for histological observation.The morphological and structural changes during vascular tissue development were systematically analyzed using light microscopy.Lignin deposition was specifically detected by combining Wiesner reaction and autofluorescence methods,while cellulose was visualized and analyzed using calcofluor white staining.[Result]The development of vascular tissues in C.cassia begins with meristematic cells at the shoot apex,which gradually differentiate into procambium initial cells.These initial cells further develop into procambium cells and phloem fibers.Some procambium cells differentiate into primary phloem and primary xylem,while others gradually develop into the cambium,which subsequently differentiates into secondary phloem and secondary xylem.During secondary growth,active periclinal divisions were observed in phloem ray cells,suggesting their possible involvement in the formation of secondary phloem cells.As vascular tissues develop,phloem fiber cells undergo cell wall thickening and exhibit relatively high lignin and cellulose contents.Xylem consistently maintains a high lignin content,while cambium cells contain almost no lignin and have a lower cellulose content compared with xylem and phloem.[Conclusion]This study provides a clear depiction of the developmental trajectories of both primary and secondary vascular tissues in C.cassia,along with the dynamic changes and characteristics of lignin and cellulose deposition in the cell walls of these tissues.These findings lay a foundation for further investigation into the regulatory mechanisms underlying vascular tissue development in C.cassia and offer valuable insights for future studies on the quality formation mechanism of this medicinal plant.
