基于转录组学探究金鱼早期发育过程中脂代谢相关基因的表达OA
Exploring lipid metabolism-related gene expression during early goldfish development via transcriptomics
为探究金鱼(Carassius auratus)早期发育过程中脂代谢相关基因的表达情况,本研究通过转录组测序分析了金鱼2种品系(鹤顶红与珍珠)早期发育3个时间点(0 h、48 h、5 d)的差异表达基因,并进行了qRT-PCR验证.结果表明,2种金鱼早期发育的3个时期均存在大量的差异表达基因.富集分析显示,2种金鱼48 h vs 0 h组、5 d vs 48 h组、5 d vs 0 h组下调差异基因主要富集在RNA转运、嘧啶代谢、卵母细胞减数分裂等通路,但上调差异基因仅在5 d vs 48 h存在富集的通路,且2种金鱼均富集在PPAR信号通路.根据富集分析结果,推测apoba、fads2、pparα、pparγ等12个关键基因参与金鱼早期发育过程中的脂质代谢调控.综上,2种金鱼5 d和48 h 2个发育时期均在PPAR信号通路及脂代谢相关基因表达水平上呈现显著差异,证实金鱼出膜后脂质代谢能力显著增强.
To elucidate the expression dynamics of lipid metabolism-related genes during early development in goldfish(Carassius auratus),we conducted transcriptome sequencing to profile differentially expressed genes(DEGs)at three critical developmental stages(0 h,48 h,and 5 d post-fertilization)in two distinct goldfish varieties(Oranda and Pearl-scale).Subsequent qRT-PCR validation confirmed the transcriptomic findings.Comparative analysis revealed a significant number of DEGs across all developmental transitions in both varieties.Enrichment analysis indicated that downregulated DEGs(48 h vs 0 h,5 d vs 48 h,and 5 d vs 0 h)were predominantly associated with pathways involved in RNA trans-port,pyrimidine metabolism,and oocyte meiosis.In contrast,upregulated DEGs exhibited pathway enrichment exclu-sively in the 5 d vs 48 h comparison,with both varieties showing significant enrichment in the PPAR signaling pathway.Further analysis identified twelve candidate genes including apoba,fads2,pparα,and pparγ as potential key regulators of lipid metabolism during early goldfish development.Collectively,these findings demonstrate pronounced differences in PPAR signaling and lipid metabolism-related gene expression between the 5 d and 48 h stages,supporting the hypothesis that post-hatching goldfish undergo a marked enhancement in lipid metabolic capacity.
