副溶血弧菌噬菌体VpP2326的分离鉴定及表征OA
Isolation,identification and characterization of a Vibrio parahaemolyticus phage VpP2326
为促进基于噬菌体的副溶血弧菌防控技术开发,以副溶血弧菌RIMD2210633菌株为宿主菌,采用双层琼脂平板法分离纯化噬菌体,并探究其生物学特性.结果表明:分离的噬菌体VpP2326头部呈二十面体结构,尾部较短.该噬菌体对宿主菌具有显著的抑菌效果,且具有较强的宿主特异性.其最佳感染复数为0.1,潜伏期小于5 min,裂解量约为147 PFU·cell-1.VpP2326具有较好的热稳定性和pH稳定性,对氯仿和蛋白酶敏感,对紫外辐照高度敏感.其核酸类型为双链DNA,基因组全长约为43 kb,G+C含量为49.37%,基因组中预测有46个开放阅读框.综上,该研究为噬菌体VpP2326的开发应用奠定了基础.
To promote the development of phage-based technologies for the prevention and control of Vibrio parahae-molyticus,a phage was isolated and purified by the double-layer agar plate method with V.parahaemolyticus RIMD2210633 strain as the host bacterium.Thereafter,its biological characteristics were explored.The results showed that the isolated phage VpP2326 had an icosahedral head and a short tail.It exhibited a significant antibacterial effect on the host bacterium and a strong host specificity.The optimal multiplicity of infection(MOI)of this phage was 0.1,the incubation period was less than 5 min,and the burst size was approximately 147 PFU·cell-1.VpP2326 had good thermal and pH stabili-ty,was sensitive to chloroform and proteases,and was highly sensitive to UV radiation.The nucleic acid type of this phage was double-stranded DNA,the whole genome was approximately 43 kb,with the G+C content of 49.37%,46 open reading frames are predicted in the genome.This study laid the foundation for the development and application of VpP2326.
