基于cAMP/PKA/CREB通路探讨红花化瘀汤减轻肌筋膜疼痛综合征模型大鼠炎性损伤作用及机制OA
Effects and Mechanisms of Honghua Huayu Decoction in Alleviating Inflammatory Damage in Rats with Myofascial Pain Syndrome via cAMP/PKA/CREB Pathway
目的:探究红花化瘀汤减轻肌筋膜疼痛综合征(Myofascial pain syndrome,MPS)炎性损伤的机制.方法:将雄性SD大鼠随机分为对照组、模型组、塞来昔布组、红花化瘀汤组、红花化瘀汤+蛋白激酶A(PKA)抑制剂H-89组(红花化瘀+H-89组).采用肌电图仪检测自发电活动频率,爪压力测痛仪检测激痛点压痛阈值,酶联免疫吸附试验法(ELISA)检测血清中P物质(SP)、降钙素基因相关肽(CGRP)、白介素-1β(IL-1β)、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、白介素-10(IL-10)水平,HE染色检测肌肉组织病理学改变,Western blot检测肌肉组织中分化簇68(CD68)、分化簇206(CD206)蛋白表达及磷酸化蛋白激酶A(p-PKA)/PKA、磷酸化cAMP反应元件结合蛋白(p-CREB)/cAMP反应元件蛋白(CREB)比值,ELISA检测肌肉组织中环磷酸腺苷(cAMP)水平.结果:相对于对照组,模型组自发电位活动频率、SP、CGRP、CD68、IL-1β、IL-6、TNF-α升高(P<0.05),激痛点压痛阈值、CD206、IL-10、cAMP、p-PKA/PKA、p-CREB/CREB降低(P<0.05),肌肉组织发生病理学改变;塞来昔布及红花化瘀汤均能改善MPS大鼠的上述病理学指标(P<0.05);红花化瘀汤的干预效果优于塞来昔布组(P<0.05);H-89能部分逆转红花化瘀汤的作用(P<0.05).结论:红花化瘀汤可能通过影响巨噬细胞表型相关蛋白的表达,减轻MPS大鼠的炎症及疼痛反应,其机制与激活cAMP/PKA/CREB信号通路有关.
Objective:To investigate the mechanism by which Honghua Huayu Decoction alleviates inflammatory damage in myofascial pain syndrome(MPS).Methods:Male SD rats were randomly divided into control group,model group,celecoxib group,HHD group,and HHD plus protein kinase A(PKA)inhibitor H-89 group.An electromyograph was used to measure spontaneous motor activity frequency;a paw pressure analgesia meter to assess the pain threshold at trigger points;ELISA to detect serum levels of substance P(SP),calcitonin gene-related peptide(CGRP),interleukin-1 β(IL-1β),interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),and interleukin-10(IL-10)levels;HE staining to detect pathological changes in muscle tissue;Western blot to detect the expression of CD68 and CD206 proteins in muscle tissue,as well as the p-PKA/PKA and p-cAMP response element-binding protein(CREB)/CREB ratios;and ELISA to detect cyclic adenosine monophosphate(cAMP)levels in muscle tissue.Results:Compared with the control group,the model group exhibited increased spontaneous electrical activity frequency,SP,CGRP,CD68,IL-1β,IL-6,and TNF-α(P<0.05),decreased trigger point pain thresholds,CD206,IL-10,cAMP,p-PKA/PKA,and p-CREB/CREB(P<0.05),and pathological changes occurred in the muscle tissue;both celecoxib and HHD significantly improved the aforementioned pathological indicators in rats with MPS(P<0.05);the therapeutic effect of HHD was superior to that of the celecoxib group(P<0.05);H-89 partially reversed the effects of HHD(P<0.05).Conclusion:Honghua Hua;Decoctic may alleviate inflammatory and pain responses in rats MPS by influencing the expression of proteins associated with macrophage phenotypes,and its mechanism is related to the activation of the cAMP/PKA/CREB signaling pathway.
